摘要:
目的观察抗疏强骨颗粒对去卵巢模型大鼠血清碱性磷酸酶(alkaline phosphatase,ALP)、抗酒石酸酸性磷酸酶5b(tartrate resistant acid phosphatase 5b,TRACP5b)的影响及其可能作用机理。 方法将SD大鼠100只随机分为正常组、模型组、中药(大、中、小剂量)组,每组20只。除正常组外其余动物饲养1周去除卵巢建立骨质疏松大鼠模型,术后1周中药组按照相应剂量给药。观察各组大鼠灌胃前后一般情况、股骨组织病理学改变以及血清ALP、TRACP5b的表达情况。 结果模型组大鼠精神状况较差,毛发颜色晦暗无光泽,体质量下降显著,偶有毛发脱落等;中药组大鼠精神状况尚可,毛发颜色较正常组稍差,体质量轻微下降。HE染色显示模型组大鼠软骨细胞出现量空骨陷窝,细胞核聚缩,且排列不整齐,骨小梁内骨细胞明显减少;中药组软骨细胞排列尚规则,结构尚完整,空骨陷窝较少见,与正常组接近。治疗3月后与模型组相比,中药组大鼠血清ALP、TRACP5b含量均下降,中剂量组含量最低,差异具有统计学意义(P<0.05)。 结论抗疏强骨颗粒能通过降低去卵巢模型大鼠血清ALP、TRACP5b表达,降低破骨细胞活性,抑制骨吸收,从而治疗绝经后骨质疏松症。
Abstract:
ObjectiveTo observe the effects of anti-osteoporosis strengthening bone granules on ALP and TRACP5b in ovariectomized rats and its possible mechanism. MethodsAll 100 SD rats were randomized into the normal group, the model group, different dosages groups of materia medica (large, moderate and small), 20 rats each group. Except the normal group, the rest of the animals were raised for one week, and the ovaries were removed to establish the osteoporosis rat model, one week after operation, materia medica groups were given the corresponding dosages of herbs. To observe general situations before and after gavage, histopathological changes of femur, the expressions of ALP and TRACP5b of the rats in different groups. ResultsThe mental state of the rats in the model group was poor, the hair color was dull, the body mass decreased significantly, and the hair fell off occasionally; the mental state of the rats in materia medica group was fair, the hair color was slightly worse than that in the normal group, and the body weight decreased slightly. HE staining showed that the chondrocytes in the model group had empty bone lacunae, nuclear condensation and irregular arrangement, and the number of osteocytes in the trabecular bone decreased significantly; the arrangement of chondrocytes in materia medica group was regular, the structure was complete, empty bone lacuna was rare, which was close to the normal group. Compared with the model group, the levels of serum ALP and TRACP5b in materia medica group decreased after three months of treatment, and the level in the moderate dose group was the lowest, the difference had statistical meaning (P<0.05). ConclusionAnti-osteoporosis strengthening bone granules could treat postmenopausal osteoporosis through lowering the expressions of ALP and TRACP5b, reducing the activity of osteoclasts and inhibiting bone resorption.
