摘要:
目的探讨四君子汤治疗结直肠癌的作用机制。 方法以网络拓扑参数筛选方法构建四君子汤治疗结、直肠癌的化合物-靶标核心网络;使用网络稳定性测试的方法寻找四君子汤治疗结直肠癌的核心化合物。 结果得到包含化合物7个及靶标33个的四君子汤治疗结直肠癌化合物-靶标核心网络;使用鲁棒性进行网络稳定性检测,得到四君子汤核心化合物4种,包括高丽槐素、罗通定、山萘酚和常春藤皂苷元;富集分析发现4种化合物与四君子汤强关联PI3K-Akt信号通路、cGMP-PKG信号通路及PPAR信号通路。 结论四君子汤治疗结、直肠癌的潜在作用机制可能是通过调控PI3K-Akt、cGMP-PKG和PPAR信号通路及药物结合、质膜区响应和肠神经元,进而影响脂代谢、细胞生存、细胞周期与细胞凋亡。
Abstract:
ObjectiveTo explore the mechanism of Sijunzi decoction in treating colorectal cancer. MethodsNetwork topology parameter screening method was used to construct compound target core network of Sijunzi decoction in the treatment of colorectal cancer; network stability test was used to survey the core compounds of Sijunzi decoction in treating colorectal cancer. ResultsThe compound target core network of Sijunzi decoction in treating colorectal cancer was obtained,containing seven compounds and 33 targets;robustness was used to perform network stability detection,and four core compounds of Sijunzi decoction were obtained,including sophorae,Rotundine,kaempferol and ivy sapogenin,enrichment analysis found that four compounds and Sijunzi decoction were strongly associated with PI3K Akt signaling pathway,cGMP PKG signaling pathway and PPAR signaling pathway. ConclusionThe potential mechanism of Sijunzi decoction in the treatment of colorectal cancer is related to the regulation of PI3K-Akt,cGMP-PKG and PPAR signaling pathways,drug binding, plasma membrane response and intestinal neurons, thereby affecting lipid metabolism, cell survival, cell cycle and apoptosis.
