摘要:
目的探讨抵当汤早期干预对2型糖尿病大鼠血管内皮细胞(vascular endothelial cells,VEC)黏附连接作用的影响。 方法采用投喂高脂饲料并尾静脉注射链脲佐菌素(streptozotocin,STZ)的方法建立2型糖尿病大鼠模型,将大鼠随机分为7组(正常对照组、糖尿病模型组、二甲双胍组、辛伐他汀组及抵当汤早、中、晚期干预组),采用Western-Blot法检测各组大鼠VEC钙黏蛋白(VE-Cad)表达;实时荧光定量PCR方法检测Ras相关蛋白1(RAS-related protein 1,Rap1)、膜相关鸟苷酸激酶1(MAGI1)、酪氨酸蛋白激酶2(Tie2)的表达水平;HE染色后观察各组大鼠胸主动脉结构。 结果与正常对照组比较,糖尿病模型组VE-Cad、Rap1、MAGI1、Tie2表达下降(P<0.05);与糖尿病模型组比较,抵当汤早、中期干预组VE-Cad、Rap1、MAGI1、Tie2表达上升(P<0.05);与抵当汤晚期干预组比较,早期干预组VE-Cad、Rap1、MAGI1、Tie2表达上升(P<0.05)。 结论抵当汤早期干预可增强VEC间黏附连接,增强血管稳定性,改善血管通透性,延缓糖尿病大血管病变的发展。
Abstract:
ObjectiveTo explore the effects of early intervention of Didang decoction on the adhesion junction of VEC in T2DM rats. MethodsRat models with T2DM were established by feeding high fat feed and injecting STZ into caudal vein, randomized into seven groups (normal control group, DM model group, metformin group, simvastatin group, early, middle and late intervention groups of Didang decoction, Western blot method was adopted to detect the expressions of VE-Cad, real time fluorescence quantitative PCR to measure the expressions of Rap1, MAGI1 and Tie2; to observe thoracic aortic structure after HE staining. ResultsCompared with normal control group, the expressions of VE-Cad, Rap1, MAGI1 and Tie2 reduced in DM model group (P<0.05); compared with DM model group, the expressions of VE-Cad, Rap1, MAGI1 and Tie2 increased in early and middle intervention groups of Didang decoction (P<0.05); compared with late intervention group of Didang decoction, the expressions of VE-Cad, Rap1, MAGI1 and Tie2 rosed in early intervention group of Didang decoction (P<0.05). ConclusionEarly intervention of Didang decoction could enhance the adhesion junction of VEC, strengthen vascular stability, improve vascular permeability and delay the development of diabetic macroangiopathy.
