目的探讨芪麝丸对背根节神经元环氧合酶2-前列腺素E2受体(cyclooxygenase 2-prostaglandin E2-E-prostanoid,COX2-PGE2-EP)信号通路相关基因和蛋白表达的影响。 方法体外分离并培养大鼠背根节神经元,将其分为对照组、模型组、芪麝丸组。培养7天后分别检测环氧合酶2的信使核糖核酸(cyclooxygenase 2 messenger RNA,COX2 mRNA)、前列腺素E2的信使核糖核酸(prostaglandin E2 messenger RNA,PGE2 mRNA)、前列腺素E2受体的信使核糖核酸(E-prostanoid Messenger RNA,EP mRNA)及环氧合酶2(cyclooxygenase,COX2)蛋白、前列腺素E2受体4(E-prostanoid 4,EP4)蛋白的表达水平。 结果模型组中加入肿瘤坏死因子α(tumor necrosis factor α,TNF-α)后COX2 mRNA、EP2 mRNA、EP4 mRNA表达上调,芪麝丸组各指标表达水平下降;TNF-α能够促进COX2蛋白及EP4蛋白表达增加,芪麝丸组各蛋白表达水平均下调。 结论芪麝丸可能下调离体培养的背根节神经元中COX2-PGE2-EP信号通路的表达,抑制背根节痛觉的表达。
Abstract:
ObjectiveTo explore the influence of Qishe pills on the expressions of COX2-PGE2-EP signaling pathway related gene and protein in dorsal root ganglion neurons. MethodsDorsal root ganglion neurons of the rats were isolated and cultured in vitro, randomized into the control group, the model group and Qishe pills group. To detect the expressions of COX2 mRNA, PGE2 mRNA, EP mRNA, COX2 protein and EP4 protein respectively after culturing for seven days. ResultsThe expressions of COX2 mRNA, EP2 mRNA and EP4 mRNA of the model group up-regulated after adding TNF-α, the expressions of the indexes in Qishe pills group down-regulated; TNF-α could increase the expressions of COX2 protein and EP4 protein, the expressions of the proteins in Qishe pills group reduced. ConclusionQishe pills may down regulate the expressions of COX2-PGE2-EP signaling pathway in cultured dorsal root ganglion neurons and inhibit the expressions of dorsal root ganglion pain.
