目的探讨龙葵总碱对人多发性骨髓瘤细胞系RPMI8226荷瘤裸鼠的抑瘤作用,观察透射电镜下细胞超微结构变化,探索龙葵总碱抗骨髓瘤的相关机制。 方法传代培养人骨髓瘤细胞RPMI8226,将90只裸鼠随机分为空白组、模型组、硼替佐米组及龙葵总碱低、中、高剂量组,每组15只。除空白组外,其余各组采用刘瑜实验法制备多发性骨髓瘤小鼠模型,造模成功后连续分别灌胃龙葵总碱及皮下注射硼替佐米各14天,断颈处死裸鼠,取出瘤体组织,根据肿瘤质量计算抑瘤率。并进行组织细胞形态观察和透射电镜检测。 结果龙葵总碱能抑制人骨髓瘤细胞的增殖;龙葵总碱低、中、高剂量组和硼替佐米组裸鼠瘤重及瘤体积较对照组减轻、变小(P<0.05);透射电镜下不同剂量龙葵总碱干预的荷瘤裸鼠均出现胞质空泡样变,核固缩以及明显的凋亡小体,自噬小体数量明显升高,自噬活性可能被上调。 结论龙葵总碱对人多发性骨髓瘤荷瘤裸鼠具有抑瘤作用,其机制可能为龙葵总碱引起的强烈自噬减弱了骨髓瘤细胞的生存力。
Abstract:
ObjectiveTo explore the inhibitory effects of total alkaloids of solanum nigrum (TASN) on human myeloma RPMI8226 tumor bearing nude mice, observe ultrastructural changes of cells under transmission electron microscope and survey the related mechanism of TASN against myeloma. MethodsAfter culturing human myeloma cells RPMI8226, 90 nude mice were chosen and randomized into the blank group, the model group, bortezomib group, low, moderate and high dosages groups of TASN, 15 rats in each group. Except the blank group, the mice in the other groups were prepared into the models of multiple myeloma using Liu Yu’s experimental method, after successfully modeling, the mice were drenched with TASN and accepted subcutaneous injection of bortezomib for 14 consecutive days respectively, the nude mice were killed by neck amputation, the tumor tissues were taken out, and the tumor inhibition rate was calculated according to the tumor mass. The morphology of tissues and cells was observed and detected by transmission electron microscope. ResultsTASN could inhibit the proliferation of human myeloma cells; tumor weight and tumor volume in nude mice of low, moderate and high dosages groups of TASN and bortezomib group reduced than these of the control group (P<0.05); there were cytoplasmic vacuolar degeneration, nuclear pyknosis and obvious apoptotic bodies in all tissues and cells in different dosages groups of TASN, the number of autophagosomes increased significantly, autophagy activity may be up-regulated. ConclusionTASN has antitumor effects on human multiple myeloma bearing nude mice, and its mechanism might be that the strong autophagy caused by TASN weakens the viability of myeloma cells.
