目的建立白芍生品中没食子酸、芍药苷及芍药内酯苷含量的快速测定近红外光谱定量模型。 方法以HPLC检测值为参照,采用傅里叶变换近红外漫反射光谱技术采集白芍生品的近红外光谱,结合偏最小二乘法(PLS)分别建立没食子酸、芍药苷及芍药内酯苷含量的快速测定方法。 结果白芍中没食子酸、芍药苷及芍药内酯苷的校正模型相关系数(R)、校正均方差(RMSEC)、内部验证均方差(RMSEP)分别为0.99132、0.0055、0.00896;0.99869、0.0266、0.10900;0.97052、0.0349、0.04210。通过外部验证,白芍的预测值与真实值相关系数分别为0.9404、0.9412和0.9446。 结论该方法准确、快速、简便,可实现大批量样品的快速分析。
Abstract:
ObjectiveTo establish a quantitative model for the rapid determination of gallic acid, paeoniflorin and albiflorin in raw Baishao (Radix Paeoniae Alba) by near-infrared spectroscopy. MethodsBy taking the detection value of HPLC as the reference, Fourier transform near infrared diffuse reflectance spectroscopy was adopted to collect near-infrared spectroscopy of raw Baishao, the method for rapid determination of the contents of gallic acid, paeoniflorin and albiflorin was established by partial least squares (PLS). ResultsCorrection model correlation coefficient (R), corrected mean square error deviation (RMSEC) and mean square error deviation of internal validation (RMSEP) of gallic acid, paeoniflorin and albiflorin in Baishao were 0.99132, 0.0055 and 0.00896; 0.99869, 0.0266 and 0.10900; 0.97052, 0.0349 and 0.04210 respectively. Correlation coefficients between predicted value and real value of Baishao reached 0.9404, 0.9412 and 0.9446 by external verification. ConclusionThe method, accurate, rapid and simple, could realize the rapid analysis of the samples in large scales.
