摘要:
目的观察抗病毒口服液对体内外感染甲型H1N1流感病毒模型的防治作用。 方法体内实验采用甲型H1N1流感病毒FM1及PR8株经滴鼻感染的70只ICR小鼠来制备肺炎模型,随机分为正常组,模型组,利巴韦林组,双黄连组,抗病毒口服液2倍量、1倍量、1/2倍量组,每组10只。经预防给药后,计算各组小鼠肺指数;经治疗给药后计算各组小鼠死亡率和平均存活时间。体外采用甲型H1N1流感病毒及H3N2病毒感染MDCK细胞致细胞病变CPE模型,经治疗给药后,计算半数抑制浓度(median inhibition concentration,IC50)及治疗指数(therapeutic index,TI)。 结果体内实验中,抗病毒口服液各剂量组与模型组、正常组比较,能明显降低流感病毒感染后小鼠肺指数,降低死亡率,延长平均存活时间(P<0.05)。体外感染实验中,抗病毒口服液给药后可明显降低病毒感染后细胞的病变情况。 结论抗病毒口服液对甲型H1N1流感病毒体内外感染有较好的治疗和预防作用。
Abstract:
ObjectiveTo observe the preventive and therapeutic effects of Kangbingdu oral liquid on influ-enza A (H1N1) virus model in vivo and in vitro. MethodsSeventy ICR mice models were infected with influenza A H1N1 virus FM1 and PR8 strains by intranasal drip in vitro to prepare pneumonia model, and randomized into the normal group, the model group, ribavirin group, Shuanghuanglian group, two-fold, one-fold and 1/2-fold dose groups of Kangbingdu oral liquid, ten rats in each group, after preventive administration, to calculate lung index in different groups, to calculate the mortality rate and mean survival time of the mice in different groups after therapeutic administration. In vitro, MDCK cells were infected with influenza A (H1N1) virus and H3N2 virus to induce CPE model, after therapeutic administration, to calculate IC50 and TI. ResultsIn in-vivo experiment, compared with the model group and the normal group, different doses of Kangbingdu oral liquid could notably lower lung index of the mice after H1N1 infection, reduce mortality rate and prolong average survival days (P<0.05); in in-vitro experiment, Kangbingdu oral liquid could apparently reduce the pathological changes of cells after virus infection. ConclusionKangbingdu oral liquid has good therapeutic and preventive effects on influenza A (H1N1) virus infection in vivo and in vitro.
