目的建立一测多评法(quantitative analysis of multi-components by single-marker,QAMS)同时测定白芍中没食子酸(A)、儿茶素(B)、芍药苷内酯(C)、芍药苷(S)、1,2,3,4,6-五没食子酰基葡萄糖(D)、苯甲酰芍药苷(E)6种成分的含量。 方法采用HPLC法,Shim-pack GIST-HP C18色谱柱(250 mm×4.6 mm,5.0 μm),乙腈(A)-0.1%磷酸水溶液(B)为流动相,梯度洗脱,流速1 mL/min,柱温30℃,检测波长232 nm。以芍药苷为内参物,建立没食子酸、儿茶素、芍药苷内酯、1,2,3,4,6-五没食子酰基葡萄糖、苯甲酰芍药苷与内参物芍药苷的相对校正因子(f),采用QAMS与外标法计算白芍中6种成分的含量,比较分析两种方法计算结果是否存在差异。 结果没食子酸、儿茶素、芍药苷内酯、芍药苷、1,2,3,4,6-五没食子酰基葡萄糖、苯甲酰芍药苷质量浓度分别在5.27~105.31 μg/mL(r=0.999 3)、5.77~115.43 μg/mL(r=0.999 0)、20.16~403.12 μg/mL (r=0.999 3)、49.87~997.35 μg/mL(r=0.999 7)、15.18~303.54 μg/mL(r=0.999 4)、5.37~107.35 μg/mL (r=0.999 0)范围内线性关系良好;加样回收率为98.6%~99.4%;以芍药苷为内参物,测得f值分别为:fA/S=0.699、fB/S=0.548、fC/S=1.470、fD/S=0.709及fE/S=0.656,建立的QAMS法计算结果与外标法计算结果比较无显著差异。 结论本实验建立的QAMS法同时测定白芍中6种成分方法简单、可靠、结果准确,可用于白芍多指标质量控制。
Abstract:
ObjectiveTo establish the method of determining six components including gallic acid (A), catechin (B), paeoniflorin lactone (C), paeoniflorin (S), 1, 2, 3, 4, 6-pentagalloyl glucose (D) and benzoyl paeoniflorin (E) in Baishao (paeoniae radix alba) by quantitative analysis of multi-components by single-marker (QAMS). MethodsHPLC was adopted, Shim-pack GIST-HP C18 chromatographic column (250 mm×4.6 mm, 5.0 μm), acetonitrile(A)-0.1% phosphoric acid aqueous solution (B) as the mobile phase, gradient elution, flow rate 1mL/min, column temperature 30℃, detection wavelength was 232 nm. By taking paeoniflorin as internal reference, the relative correction factors(f) between A, B, C, D, E and S were established, the contents of six components in Baishao were calculated by using QAMS and external standard method, to compare and analyze whether there are differences between the calculation results of the two methods. ResultsThe concentrations of A, B, C, S, D and E presented the good linear relationship in the range from 5.27 to 105.31 μg/mL (r=0.999 3), 5.77 to 115.43 μg/mL (r=0.999 0), 20.16 to 403.12 μg/mL (r=0.999 3), 49.87 to 997.35 μg/mL (r=0.999 7), 15.18 to 303.54 μg/mL (r=0.999 4), 5.37 to 107.35 μg/mL (r=0.999 0); average recovery was between 98.6% and 99.4%; by taking paeoniflorin as internal reference, the detected f values were fA/S=0.699, fB/S=0.548, fC/S=1.470, fD/S=0.709 and fE/S=0.656 respec-tively, there was no significant difference between the calculation results of QAMS and external standard method. ConclusionQAMS built in the experiment is simple, reliable and accurate, it could detect six components in Baishao simultaneously and could be used for multi-index quality control of Baishao.
