目的研究益肾固本膏方对肾阳虚哮喘模型大鼠皮质类固醇合成酶11β-羟基类固醇脱氢酶1[hydroxysteroid(11-beta)dehydrogenase 1,HSD11B1)]、11β-羟化酶1(CYP11B1)及糖皮质激素受体亚型GRα、GRβ的影响。 方法将40只大鼠随机分为空白对照组,模型对照组,益肾固本膏方低、中、高剂量组,每组8只。建立肾阳虚哮喘大鼠模型,空白对照组与模型对照组以蒸馏水2 mL灌胃,益肾固本膏方各剂量组给予对应药物剂量灌胃,各组均灌胃20天。RT-PCR法测定大鼠肾上腺组织中HSD11B1、CYP11B1 mRNA及肺组织中GRα、GRβ mRNA表达;Western Blot法测定大鼠肾上腺组织中HSD11B1、CYP11B1及肺组织中GRα、GRβ蛋白表达。 结果益肾固本膏方各剂量组HSD11B1、CYP11B1表达均高于模型组,差异有统计学意义(P<0.01)。益肾固本膏方各剂量组GRα表达均高于模型对照组(P<0.01),GRβ表达均低于模型对照组(P<0.01)。 结论益肾固本膏方可促进HSD11B1、CYP11B1合成,升高肾阳虚哮喘模型大鼠的血浆皮质酮水平,通过上调GRα含量、下调GRβ含量增强激素效应。
Abstract:
ObjectiveTo study the effects of Yishen Guben ointment (YSGB) on corticosteroid synthase 11β-hydroxysteroid dehydrogenase 1 (HSD11B1), 11β-hydroxylase 1(CYB11B1) and glucocorticoid receptor subtypes GRα and GRβ in rats with kidney-yang deficiency pattern asthma. MethodsForty rats were randomized into blank control group, model control group, low, moderate and high doses groups of YSGB, eight rats in each group. Rat models with kidney-yang deficiency pattern asthma were established, blank control group and model control group were perfused with distilled water, 2mL, different doses groups of YSGB were drenched with the correspon-ding drug dose, all groups were administered for 20 days. To detect the expressions of HSD11B1, CYP11B1 mRNA and GRα and GRβ mRNA in lung tissue by RT-PCR method; to measure the protein expression of HSD11B1, CYP11B1 in adrenal tissue and GRα and GRβ protein in lung tissue by Western Blot method. ResultsThe expressions of HSD11B1 and CYP11B1 in different doses groups of YSGB were higher than these of the model group, and the difference had statistical meaning (P<0.01). The expressions of GRα in different doses groups of YSGB were higher than these of model control group (P<0.01), the expressions of GRβ lower than these of model control group (P<0.01). ConclusionYSGB could raise the levels of plasma corticosterone in rat models with kidney-yang deficiency pattern asthma by promoting the synthesis of HSD11B1 and CYP11B1, it could enhance hormone effects by up regulating the contents of GRα and down regulating the contents of GRβ.
