目的观察四逆散对应激性抑郁模型大鼠血清白细胞介素1β(interleukin,IL-1β)、海马组织五羟色胺(5-hydroxytryptamine,5-HT)含量变化,探讨四逆散对抑郁症的影响。 方法选择50只SD大鼠,按随机数字表法分为空白组,模型组,四逆散高、低剂量组,氟西汀组,每组10只。除空白组外,其余组均采用慢性不可预性温和应激结合孤养方式建立抑郁大鼠模型28天。造模同时,四逆散各剂量组给予四逆散对应剂量灌胃,氟西汀组予盐酸氟西汀分散片溶液灌胃,空白组及模型组给予生理盐水灌胃。观察各组大鼠体质量,行为学变化、糖水偏好率;测定大鼠血清IL-1β、海马组织5-HT水平。 结果给药28天后,与模型组及氟西汀组比较,四逆散高剂量组大鼠体质量、行为学评分及糖水偏爱率均增加(P<0.05)。与空白组相比,模型组大鼠血清IL-1β升高、海马组织5-HT含量下降(P<0.05);与模型组相比,四逆散各剂量组、氟西汀组血清IL-1β下降、海马组织5-HT含量升高(P<0.05),氟西汀组作用较明显。 结论四逆散可能通过下调血清IL-1β含量,提高海马5-HT含量,改善抑郁大鼠行为,从而起到抗抑郁作用。
Abstract:
ObjectiveTo observe the changes of serum IL-1β and 5-HT in hippocampus of stress depression model rats treated by Sini powder, and to explore the intervention effect of Sini powder on depression. MethodsFifty SD rats were separated into the blank group, the model group, high and low dose groups of Sini powder, fluoxetine group, with ten rats in each group. Except the blank group, the other groups adopted chronic unpre-dictable mild stress and solitary feeding to establish the depression rat model for 28 days. Meanwhile, different doses groups of Sini powder were drenched with the corresponding dose of Sini powder, fluoxetine group with the solution of fluoxetine hydrochloride dispersible tablet, the blank group and the model group with physiological saline. To observe body mass, behavioral changes and sugar water preference rate of the rats in different groups; and to detect the levels of serum IL-1β and 5-HT in hippocampus of the rats. ResultsAfter medicating for 28 days, compared with the model group and fluoxetine group, body mass, behavioral scores and sugar water preference rate rose in high dose group of Sini powder (P<0.05). Compared with the blank group, the levels of serum IL-1β lifted and the contents of 5-HT decreased in hippocampus of the rats in the model group (P<0.05); compared with the model group, the levels of serum IL-1β decreased and the contents of 5-HT increased in hippocampus of the rats in different doses groups of Sini powder and fluoxetine group (P<0.05), the effects of fluoxetine group were more notable. ConclusionSini powder could raise the contents of 5-HT in hippocampus and improve the behaviors of depression rats possibly through down regulating the contents of serum IL-1β, thus developing anti-depression functions.
