摘要:
目的探究屏风四妙汤治疗复发性外阴阴道假丝酵母菌病小鼠模型疗效及机制。 方法选取120只雌性ICR小鼠,随机分为对照组和A、B、C、D、E、F组,每组16~18只。A、B组为雌激素化接种白色念珠菌组;C、D组为免疫抑制接种白色念珠菌组;E、F组为正常免疫组。对照组不做任何处理;A组予雌激素+中药处理10天;B组予雌激素处理1天;C组予地塞米松3次+中药处理10天;D组予塞米松处理3次;E组予植物油+中药处理10天;F组予植物油处理10天。采用免疫组织化学染色(immunohistochemistry,IHC)法检测小鼠阴道内CD11c+水平;取各组小鼠阴道组织常规处理行苏木精-伊红染色法(hematoxylin-eosin staining,HE)和碘酸雪夫染色(periodic acid-schiff stain,PAS)进行病理观察并评分。 结果与对照组比较,A、B、C、D、E、F组病理评分显著升高(P<0.05)。CD11c+免疫组化图显示,处理后C组阳性细胞数量最少。与对照组CD11c+光密度值比较,A、B、C、D、E、F组均降低,其中C组降低最明显(P<0.05)。HE及PAS染色图显示:对照组阴道黏膜无角化、溃疡、充血和炎性浸润,A、B、C、D、E、F组阴道黏膜出现不同程度的上皮角化、炎性浸润和充血情况,其中A组阴道黏膜角化及出血症状最轻。 结论灌服屏风四妙汤煎剂可以有效缓解白色念珠菌感染小鼠阴道内黏膜充血和炎细胞浸润。
Abstract:
ObjectiveTo investigate clinical effect of Pingfeng Simiao decoction in the treatment of mice models with recurrent vulvovaginal candidiasis (RVVC) and the mechanism. MethodsAll 120 female ICR mice were chosen, and randomized into the control group and the groups of A, B, C, D, E and F, with 16~18 mice in each group. Group A and group B were constructed into the groups of estrogenic inoculation of candida albicans, group C and group D were prepared into the group of immunosuppressive inoculation with candida albicans; group E and F were established into the groups of normal immunity. The control group was unhandled; group A was managed by estrogen and herb for ten days; group B by estrogen for one day; group C by dexamethasone three times and herbs for ten days; group D by dexamethasone three times; group E by plant oil and herbs for ten days, and group F by plant oil IHC was used to detect the levels of intravaginal CD11c+ in the mice; vaginal tissue was taken from the mice in different groups, and pathological observation was carried out after conventional management and performing HE staining and PAS staining, and scored. ResultsCompared with the control group, pathological scores of groups of A, B, C, D, E and F were increased notably (P<0.05). Immunohistochemical chart of CD11c+ displayed that the number of positive cells in group C was the least after handling. Compared with optical density of CD11c+ of the control group, it was lowered in the groups of A, B, C, D, E and F, among them, the decrease of group C was the most notable (P<0.05). HE staining and PAS staining showed that no keratosis of vaginal mucosa, ulcer, congestion and inflammatory infiltration occurred in the control group, while different degrees of epithelial keratosis, inflammatory infiltration and congestion manifested in vaginal mucosa in the groups of A, B, C, D, E and F, the keratosis of vaginal mucosa and congestion were the slightest in group A. ConclusionThe irrigation of Pingfeng Simiao decoction could effectively alleviate the congestion and inflam-matory infiltration in vaginal mucosa in the mice with candida albicans infection.
