摘要:
目的探讨加味过敏煎对荨麻疹小鼠血清组胺含量及组胺所致血管通透性的影响。 方法采用低分子右旋糖酐诱导的小鼠皮肤瘙痒模型,观察小鼠皮肤瘙痒情况,酶联免疫吸附法(ELISA)检测小鼠血清中组胺水平;采用伊文思兰染色法,观察加味过敏煎对组织胺引起的毛细血管通透性增加的影响。 结果与模型组相比,加味过敏煎高剂量组小鼠抓挠潜伏期明显延长,抓挠次数显著减少(P<0.05),加味过敏煎各剂量组均显著缩短了小鼠的抓挠持续时间(P<0.05)。ELISA结果显示,加味过敏煎高剂量组能明显减少小鼠血清组胺含量(P<0.05)。加味过敏煎各剂量组均能减少腹腔内渗出的伊文思蓝染料量,降低OD值(P<0.05)。 结论加味过敏煎能改善荨麻疹小鼠皮肤瘙痒情况,减少体内组胺含量,降低毛细血管通透性,且高剂量效果最好。
Abstract:
ObjectiveTo discuss the influence of modified Guomin decoction (MGD) on the contents of serum histamine and histamine-induced vascular hyperpermeability in mice with urticaria. MethodsThe mouse skin itching models induced by low molecular dextran were used to observe the skin pruritus of mice, ELISA was adopted to detect the levels of histamine in the serum of the mice; Evans blue staining was used to observe the effects of MGD on histamine-induced vascular hyperpermeability. ResultsCompared with the model group, scratch latency prolonged notably, and scratch times reduced significantly in high dose group of MGD (P<0.05), all dose groups of MGD apparently shortened the scratching duration of mice (P<0.05). ELISA results showed that high dose group of MGD could notably lower the contents of serum histamine in mice (P<0.05). Different doses groups of MGD could reduce the amount of Evans blue dye exuded from abdominal cavity, lower OD values (P<0.05). ConclusionMGD could improve skin itching conditions in mice with urticaria, lower the contents of histamine, reduce capillary permeability, and high dose show the best effects.
