摘要:
目的研究脊髓康对大鼠脊髓损伤区胶质瘢痕形成及硫酸软骨素蛋白聚糖(chondroitin sulfate proteoglycans,CSPG)表达的抑制作用。 方法将60只SD大鼠随机分为正常对照组、模型组、脊髓康组及激素组,每组15只。除正常对照组外,其余3组利用改良Allen法制造脊髓损伤大鼠模型,各组给予对应药物治疗。于第3、7、14天抽取样本制备切片标本,通过HE染色与EnVision法染色观察各组大鼠脊髓受损区的病理形态学改变;实时荧光定量PCR检测各组脊髓损伤区CSPG表达水平。 结果在造模后第3、7、14天,脊髓康组和激素组相比于模型组,病理图片中脊髓损伤区域内胶质瘢痕形成更少,且CSPG表达受到明显抑制(P<0.05),同时,脊髓康组相比于激素组,病理图片中脊髓损伤区域内胶质瘢痕形成差异不明显,但CSPG表达量高于激素组(P<0.05)。 结论中药脊髓康可以抑制大鼠脊髓损伤区胶质瘢痕过度形成及CSPG的表达,对于促进脊髓损伤后神经修复再生具有积极意义。
Abstract:
ObjectiveTo study the inhibitory effects of Jisuikang on gelatinous scar formation and the expressions of chondroitin sulfate proteoglycans (CSPG) in rats after spinal cord injury. MethodsSixty SD rats were randomized into normal control group, the model group, Jisuikang group and hormone group, with 15 rats in each group. Except normal control group, the other three groups were established into rat models by using imp-roved Allen method, different groups were treated by the corresponding medicine. Section preparation was performed after samples were selected at the third, seventh and fourteenth day, pathological morphological changes in the area of spinal cord injury of the rats were observed in different groups after HE staining and EnVision method; the expression of CSPG in the area of spinal cord injury were measured by using real-time fluorescence quantitative PCR. ResultsAt the third, seventh and 14th day after modeling, the formation of gelatinous scar was less in the area of spinal cord injury in the pathological images, and the expressions of CSPG were inhibited obviously when Jisuikang group and hormone group were compared with the model group (P<0.05), meanwhile, the difference was insignificant in the formation of gelatinous scar in the area of spinal cord injury in the pathological images when Jisuikang group was compared with hormone group, while the expressions of CSPG were higher than these of hormone group (P<0.05). ConclusionJisuikang could control excessive formation of gelatinous scar and the expressions of CSPG in rats after spinal cord injury, which has a positive meaning for promoting nerve repair regeneration after spinal cord injury.
