摘要:
目的采用响应曲面法优化小叶红叶藤挥发油包合工艺,并对其包合物进行表征。 方法通过包合方法的考察,选择小叶红叶藤挥发油包合物的最佳制备方法;以挥发油包合率和包合物收率为评价指标,采用响应曲面法考察β-环糊精(β-cyclodextrin,β-CD)和小叶红叶藤挥发油投料比、包合温度、包合时间对包合工艺的影响,优化其包合工艺;通过显微鉴别、薄层色谱、紫外分光光度法和红外光谱法对包合物进行表征。 结果最佳包合工艺为β-CD和挥发油投料比为11.88 g/mL,包合温度为52.8 ℃,包合时间为1.72 h,综合评分为96.53分,包合物的预测值和理论值偏差较小,挥发油和β-CD已形成包合物。 结论响应曲面法适用于优化小叶红叶藤挥发油包合工艺。
Abstract:
ObjectiveTo optimize the inclusion process of volatile oil of Xiaoye Hongyeteng (Rourea microphylla) by response surface method and characterize its clathrate compound. MethodsThrough investigating the inclusion method, the best preparation method of volatile oil clathrate of Xiaoye Hongyeteng was selected; by taking the inclusion rate of volatile oil and the yield of clathrate compound as the evaluation indexes, response surface method was adopted to survey the rate of charge between β-CD and Xiaoye Hongyeteng, the inclusion temperature and the influence of inclusion time on the inclusion process, therefore to optimize the inclusion process; microscopic identification, thin layer chromatography, ultraviolet spectrophotometry and infrared spectroscopy were used to characterize inclusion complex. ResultsThe best inclusion process is:the rate of charge between β-CD and Xiaoye Hongyeteng is 11.88 g/mL, the inclusion temperature 52.8 ℃, inclusion time 1.72 h, comprehensive scores 96.53 points, the deviation between the predicted and theoretical values of inclusion complex is small, and the inclusion complex has been formed by volatile oil and β-CD. ConclusionResponse surface method applies to the optimization of inclusion process of volatile oil of Xiaoye Hongyeteng.
