摘要:
目的探讨复方雷公藤汤最佳配比剂量正交试验设计及其对雌性肾病大鼠生殖系统的减毒增效作用。 方法采用一次性尾静脉注射阿霉素方法复制肾病模型,将雌性肾病模型大鼠随机分为9组各10只,依据正交试验设计连续灌服不同剂量配比的复方雷公藤汤14天。观察大鼠24 h尿蛋白定量及血清雌二醇(estradiol,E2)、促卵泡刺激素(follicle stimulating hormone,FSH)、促黄体生成素(luteinizing hormone,LH)水平。筛选出最佳剂量配比组合复方雷公藤汤。 结果影响生殖系统因素大小依次为雷公藤>当归>甘草>制何首乌;影响24 h尿蛋白定量因素大小依次为雷公藤>甘草>制何首乌,当归无影响。 结论雷公藤60 g、制何首乌54 g、当归72 g、甘草12 g剂量配比以减低肾病大鼠生殖系统毒性为主,雷公藤120 g、制何首乌36 g、当归36 g、甘草12 g剂量配比以降低肾病大鼠24 h尿蛋白为主,两种配比复方均起减毒增效作用。
Abstract:
ObjectiveTo discuss orthogonal design of the optimal dosage ratio of compound Leigongteng Tang and its effects on reproductive system in female rats with nephropathy. MethodsThe models with nephropathy were duplicated by injecting one shot of adriamycin through caudal vein, female models were randomized into nine groups, with ten in each group, and they were drenched with different dosage ratio of compound Leigongteng Tang for 14 days consecutively according to orthogonal design. To observe 24 h urinary protein quantitation, the levels of E2, FSH and LH, consequently to screen out the optimal dosage ratio of compound Leigongteng Tang. ResultsThe factors affecting the reproductive system ranked in the order: Leigongteng (Tripterygium wilfordii Hook.f.) >Danggui (Angelicae sinensis radix)>Gancao (Glycyrrhiza Radix ET Rhizoma) >prepared Heshouwu (Polygoni multiflori radix praeparata); the factors influencing 24 h urinary protein quantitation were Leigongteng>Gancao>prepared Heshouwu, while Danggui showed no influence. ConclusionCompound Leigongteng Tang in the dosage ratio of Leigongteng 60 g, prepared Heshouwu 54 g, Danggui 72 g and Gancao 12 g focuses on reducing reproductive toxicity, while the decoction in the dosage ratio of Leigongteng 120 g, prepared Heshouwu 36 g, Danggui 36 g and Gancao 12 g could lower 24 h urinary protein quantitation mainly, the decoction in the dosage ratio could develop the effects of toxicity reducing and efficacy enhancing.
