目的建立同时测定金铃子散中延胡索乙素、黄连碱、紫堇碱、巴马汀和去氢紫堇碱含量的一测多评法(quantitative analysis of multi-components with single marker,QAMS),验证该方法在金铃子散质量控制应用中的科学性和可行性。 方法采用Agilent 5 TC-C18色谱柱(250 mm×4.6 mm,5 μm),以乙腈-0.1%磷酸水溶液为流动相,梯度洗脱,柱温30 ℃,流速1.0 mL/min,检测波长280 nm;以延胡索乙素为内参物,建立该成分与其他4种成分的相对校正因子,以相对校正因子计算各成分含量,同时借助余弦相似度评价一测多评法计算结果和外标法实测值的差异。 结果5种成分在各自范围内线性关系良好,平均加样回收率在96.79%~102.41%之间,RSD在0.45%~2.67%之间;建立的各相对校正因子耐用性良好,一测多评法和外标法含量测定结果无显著差异。 结论利用相对校正因子对金铃子散中5种生物碱含量测定可行,所建立的一测多评法可用于金铃子散的质量评价。
Abstract:
ObjectiveTo establish a method of QAMS for simultaneous determination of tetrahydropalmatine, coptisine, corydaline, palmatine, dehydrocorydaline contained in Jinlingzi powder, so as to verify the scientificity and feasibility of the method applied in the quality control of Jinlingzi powder. MethodsAgilent 5 TC-C18 chromatographic column (250 mm×4.6 mm, 5 μm) was adopted, acetonitrile-0.1% phosphoric acid aqueous solutions as the mobile phase, gradient elution, column temperature 30 ℃, flow rate 1.0 mL/min, detection wavelength 280 nm; by taking tetrahydropalmatine as the internal reference, relative correction factor between the component and other four components were established, and the contents of different components were calculated by relative correction factor, in the meantime, to assess the difference beween the calculated results of QAMS and the measured value of external reference method by using cosine similarity. ResultsFive components presented good linearity in their own ranges, average recovery rates were between 96.79% and 102.41%, RSD was in the range from 0.45% to 2.67%; The established relative correction factors have good durability, no significant difference has been found in the results of QAMS and the ones of external reference method. ConclusionIt is feasible to determine the contents of five alkaloids in Jinlingzi powder, QAMS easblished could be applied to quality evaluation of Jinlingzi powder.
