摘要:
目的探讨益气活血中药对心肌梗死后心力衰竭大鼠心肌组织中基质金属蛋白酶1(matrix metalloproteinase1,MMP-1)、Ⅲ型胶原水平的影响及改善心室重构的机制。 方法准备SPF级SD雄性大鼠40只,分为空白组、模型组、中药组和西药组。除空白组10只外,其余大鼠制备心肌梗死后心力衰竭大鼠模型。空白组和模型组大鼠每日予蒸馏水灌胃,中药组予益气活血中药煎剂灌胃,西药组予卡托普利灌胃。用药6周后超声检测大鼠心脏变化。使用实时荧光定量PCR法检测大鼠心肌组织中MMP-1及Ⅲ型胶原mRNA的表达情况,同时应用免疫组化法检测心肌组织中MMP-1和Ⅲ型胶原蛋白的表达情况。 结果与空白组对比,模型组MMP-1 mRNA及蛋白表达明显上调(P<0.05),Ⅲ型胶原mRNA及蛋白表达明显下调(P<0.05)。与模型组比较,中、西药组MMP-1 mRNA及蛋白表达明显下调(P<0.05),Ⅲ型胶原mRNA及蛋白表达明显上调(P<0.01)。西药组与中药组MMP-1 mRNA及蛋白、Ⅲ型胶原mRNA及蛋白表达比较,差异无统计学意义(P>0.05)。 结论益气活血中药可能通过降低心肌组织中MMP-1水平、提高Ⅲ型胶原水平,延缓心室重构进程,达到改善心功能,防治心肌梗死后心力衰竭的目的。
Abstract:
ObjectiveTo explore the influence of benefiting Qi herbs for activating blood circulation on the expressions of MMP-1 and type Ⅲ collagen in myocardial tissue in rats with heart failure (HF) after myocardial infarction and its mechanism of improving ventricular remodeling. MethodsForty SPF SD male rats were allocated to the blank group, the model group, materia medica group and Western medicine group. Except ten rats of the blank group, the rest were prepared into HF models after myocardial infarction. The rats in the blank group and the model group accepted gastric perfusion of distilled water every day, the ones in materia medica group were drenched with the decoction of benefiting Qi herbs for activating blood circulation, and Western medicine group accepted intragastric administration of captopril. After administration for six weeks, to detect the changes of the heart by using ultrasound. To measure the expressions of MMP-1 and type Ⅲ collagen mRNA in myocardial tissue by applying real-time PCR method, simultaneously to detect the expressions of MMP-1 and type Ⅲ collagen by using immunohistochemical method. ResultsCompared with the blank group, the expressions of MMP-1 mRNA and protein were significantly up-regulated in the model group (P<0.05), the expressions of type Ⅲ collagen mRNA and protein were noticeably down-regulated (P<0.05). Compared with the model group, the expressions of MMP-1 mRNA and protein were significantly down-regulated in materia medica group and Western medicine group (P<0.05), the expressions of type Ⅲ collagen mRNA and protein were up-regulated apparently (P<0.01). The difference had no statistical meaning in expressions of MMP-1 mRNA and protein, type Ⅲ collagen mRNA and protein between materia medica group and Western medicine group (P>0.05). ConclusionBenefiting Qi herbs for activating blood circulation could achieve the aims of improving myocardial function, preventing and controlling heart failure after myocardial infarction possibly through lowering the levels of MMP-1 and raising the levels of type Ⅲ collagen, and postponing the progress of ventricular remodeling.
