摘要:
目的基于网络药理学探讨人参抗痴呆的作用机制。 方法采用中药系统药理学数据库与分析平台(traditional Chinese medicine systems pharmacology database and analysis platform,TCMSP)检索中药有效成分和相关靶点,利用GeneCards和OMIM数据库检索痴呆的基因靶点,构建蛋白互作网络(protein-protein interaction networks,PPI),进行GO功能富集分析及KEGG通路富集分析,构建可视化的信号通路。 结果从人参中筛选出22个有效成分,76个痴呆疾病作用靶点,112条GO生物学过程,127条KEGG通路。人参抗痴呆的核心基因主要有AKT1、CASP3、JUN、MAPK8、PTGS2、IL1B、PPARG等,这些基因可通过影响肽结合、酰胺结合、G蛋白偶联胺受体活性等来调控痴呆的发生。KEGG通路主要包括刺激神经组织的中的交互通路、流体剪切应力与动脉粥样硬化通路、肿瘤坏死因子信号通路等。 结论人参可通过多成分、多靶点、多途径抗痴呆,为将来提取人参有效成分治疗痴呆病提供了理论基础。
Abstract:
ObjectiveTo discuss the mechanism of ginseng in the treatment of dementia on the foundation of network pharmacology. MethodsThe effective constituents and the relevant targets of herbs were searched by using TCMSP, the gene targets of dementia were retrieved by using GeneCards and OMIM databases, to build PPI, conduct GO function enrichment analysis and KEGG pathway enrichment analysis, and construct visual signaling pathway. ResultsAll 22 active ingredients were screened out from ginseng, involving 76 targets of dementia, 112 biological processes of GO, 127 KEGG pathways. Core genes of ginseng in improving dementia contained AKT1, CASP3, JUN, MAPK8, PTGS2, IL1B, and PPARG, and these genes adjusted the occurrence of dementia by affecting the activity of peptide binding, amide binding, G protein-coupled amine receptors. KEGG pathways mainly covered interaction pathway stimulating nervous tissue, fluid shear stress and atherosclerosis pathway, and tumor necrosis factor signaling pathway. ConclusionGinseng could improve dementia by many ingredients, targets and ways, and it could provide theoretical foundation for the extraction of the active ingredients from ginseng for dementia in the future.
