摘要:
目的建立用高效液相色谱法(high performance liquid chromatography,HPLC)测定佛甲草中δ-香树酯酮含量的方法。 方法采用Agilent ZORBAX Eclipse XDB-C8色谱柱(150×4.6 mm,5 μm),流动相为乙腈-水-磷酸-三乙胺(85∶15∶0.1∶0.1,v/v);柱温为25 ℃,流速为1.0 mL/min,检测波长为211 nm。δ-香树酯酮在浓度为0.5~40.5 μg/mL范围内峰面积与浓度呈良好的线性关系,相关系数为R2=0.9991,平均回收率为98.7%。 结果δ-香树酯酮在此色谱条件下的保留时间为26.15 min,经分析计算可得,西安交通大学药学院药圃里佛甲草中δ-香树酯酮的含量为860.5 μg/100 g生药,在秦岭采集的佛甲草中δ-香树酯酮的含量为906.8 μg/100 g生药。 结论HPLC法测得的结果准确可靠,可作为δ-香树酯酮质量控制的一种方法。
Abstract:
ObjectiveTo establish a method of determining the contents of δ-amyrone contained in Fojiacao (Sedum lineare Thunb.) by HPLC. MethodsAgilent ZORBAX Eclipse XDB-C8 chromatographic column (150×4.6 mm, 5 μm), mobile phase: acetonitrile-water-phosphate -triethylamine (85∶15∶0.1∶0.1, v/v); column temperature:25 ℃, flow rate was 1.0 mL/min, detection wavelength was 211 nm. δ-amyrone presented good linearity between peak area and concentration in the range of 0.5 and 40.5 μg/mL, the correlation coefficient was R2=0.9991, average recovery rate was 98.7%. ResultsThe retention time of δ-amyrone was 26.15 min under the chromatographic conditions, by analysis and calculation, the contents of δ-amyrone were 860.5 μg/100 g crude drug contained in Fojiacao planted in the medicinal garden of pharmacy shcool, Xi'an Jiaotong University, while its contents were 906.8 μg/100 g crude drug contained in Fojiacao gathered from Qinling mountains. ConclusionThe results are accurate and reliable obtained by HPLC, and it could be taken as a approach for quality control of δ-amyrone.
