摘要:
目的探究平肝止痫复方联合卡马西平对难治性癫痫大鼠的相关凋亡基因表达及其微结构的影响。 方法颅内注射海人酸建立难治性癫痫大鼠模型,将大鼠随机分为假手术组、模型组、卡马西平组、平肝止痫复方组、平肝止痫复方联合卡马西平组,每组6只。各组大鼠给予相应药物28天后处死取脑组织,通过透射电子显微镜观察大鼠海马区神经元超微结构,并记录大鼠癫痫发作抽搐情况;按原代大鼠海马神经元将大鼠分为对照组、模型组、溶剂含药血清组、卡马西平含药血清组,平肝止痫复方联合卡马西平含药血清组,用TUNEL法观察并测算细胞凋亡指数,Western blot法检测BCL2-Associated X,Bax(Bax)、B淋巴细胞瘤(B-cell lymphoma-2,Bcl-2)、半胱天冬氨酸蛋白酶3(cysteinyl aspartate specific proteinase,Caspase-3)表达。 结果与假手术组大鼠比较,难治性癫痫大鼠神经元凋亡比例上升,凋亡速度增快。药物干预可降低凋亡细胞比例,并抑制其凋亡速度,平肝止痫复方联合卡马西平组效果优于卡马西平组;与对照组相比,模型组大鼠细胞凋亡指数、Bax、Caspase-3蛋白表达升高,Bcl-2表达下降;与模型组比较,平肝止痫复方联合卡马西平含药血清组对神经元细胞凋亡指数、Bax、Caspase-3表达的抑制及Bcl-2表达的促进作用最明显。 结论平肝止痫复方联合卡马西平对难治性癫痫大鼠的神经元具有保护作用,可抑制其神经元的细胞凋亡。
Abstract:
ObjectiveTo explore the effects of Pinggan Zhixian compound and carbamazepine on the expressions of apoptosis related genes and its microstructure in rats with refractory epilepsy. MethodsRat models of refractory epilepsy were established by using intracranial injection of kainic acid, and the rats were divided into sham operation group, the model group, carbamazepine group, Pinggan Zhixian compound group, the group of Pinggan Zhixian compound and carbamazepine, six rats in each group. The rats in different groups were sacrificed to take out cerebral tissue after administering the corresponding drugs for 28 days, neuronal ultrastructure in the rat hippocampus was observed by using transmission electron microscope, to record seizure and convulsions of the rats in sham operation group; the rats were allocated to the control group, the model group, solvent-containing serum group, carbamazepine-containing serum group, the group of serum containing Pinggan Zhixian compound and carbamazepine according to primary rat hippocampal neurons, cellular apoptosis index was observed and calculated by using TUNEL method, the expressions of Bax, Bcl-2 and Caspase-3 were measured by using Western blot method. ResultsThe results observed under electron microscopy show that, compared with normal rats in sham operation group, a rise in the proportion of neuronal apoptosis and accelerating apoptosis speed could be found in rats with refractory epilepsy. Drug intervention could reduce the proportion of apoptotic cells, and inhibit cellular apoptosis speed, the effects of the group of serum containing Pinggan Zhixian compound and carbamazepine are better than these of carbamazepine group; compared with the control group, the apoptosis index, the expressions of Bax and Caspase-3 protein increased in the model group, the expressions of Bcl-2 lowered; compared with the model group, the group of serum containing Pinggan Zhixian compound and carbamazepine have the obvious inhibitory effects on the apoptosis index of the neuronal cells, the expressions of Bax, Caspase-3 and Bcl-2 protein. ConclusionPinggan Zhixian compound and carbamazepine could protect the neuron in the rats with refractory epilepsy, and it could restrain cellular apoptosis of the neurons.
