摘要:
目的从TRAF6/IRAK1/NF-κB信号通路角度观察安肠汤对TNBS诱导的大鼠溃疡性结肠炎(ulcerative colitis,UC)的作用及机制。 方法选择60只实验大鼠,除空白组10只外,其余各组均采用2,4,6-三硝基苯磺酸(2,4,6-trinitrobenzenesulfonic acid sol,TNBS)/乙醇灌肠法建立UC大鼠模型,造模成功大鼠分为模型组,美沙拉嗪组,安肠汤低、中、高剂量组,每组10只。空白组、模型组灌胃生理盐水,美沙拉嗪组采用美沙拉嗪混悬液灌胃,安肠汤各剂量组给予对应剂量安肠汤灌胃。给药14天后处死各组大鼠。观察各组大鼠结肠病理改变;酶联免疫吸附测定法(enzyme-linked immune sorbent assay,ELISA)检测大鼠血清肿瘤坏死因子α(tumor necrosis factor α,TNF-α)、白细胞介素1β(interleukin 1β,IL-1β)、白细胞介素6(interleukin 6,IL-6)水平;Western blot法检测结肠组织中TNF受体关联因子6重组蛋白(recombinant tnf receptor associated factor 6,TRAF6)、白细胞介素1受体相关激酶(recombinant interleukin 1 receptor associated kinase 1,IRAK1)、磷酸化细胞核因子NF-κB抗体(phosphorylated nuclear factor NF-κB antibody,p-NF-κB)、核因子κB(nuclear factor kappa-B,NF-κB)蛋白的表达;通过定量聚合酶链反应测定大鼠结肠组织中TRAF6、IRAK1、NF-κB mRNA的相对表达量。 结果与空白组比较,模型组结肠黏膜病理受损严重,血清TNF-α、IL-1β、IL-6含量显著升高,结肠组织中TRAF6、IRAK1、p-NF-κB、NF-κB蛋白表达上调(P<0.05)。与模型组相比,安肠汤各剂量组和美沙拉嗪组大鼠血清TNF-α、IL-1β、IL-6含量显著降低(P<0.05);结肠组织中TRAF6、IRAK1、p-NF-κB、NF-κB蛋白表达下调(P<0.05)。 结论安肠汤对TNBS诱导的UC大鼠有治疗作用,其作用可能与其调节TRAF6/IRAK1/NF-κB通路及相关炎症因子,从而减轻肠道炎症反应,缓解肠黏膜损伤有关。
Abstract:
ObjectiveTo observe the effects of Anchang decoction on TNBS-induced UC rats via TRAF6/IRAK1/NF-κB signaling pathway and the mechanism. MethodsSixty Rats were chosen, except ten rats of the blank group, UC rat models were established by 2, 4, 6-trinitrobenzene sulfonic acid (TNBS)/ethanol enema in the other groups, the rats were divided into the model group, mesalazine group, low, moderate and high dose groups of Anchang decoction with ten rats in each group after modeling successfully. The blank group and the model group were drenched with physiological saline, and mesalazine group with mesalazine suspension, different dosages groups of Anchang decoction with different dosages of Anchang decoction. To sacrifice the rats in different groups after medicating for 14 days. To observe pathological changes of colon in the rats of different groups; ELISA method was used to detect the levels of TNF-α, IL-1β and IL-6 in the serum of the rats; Western blot method to measure the expressions of TRAF6, IRAK1, p-NF-κB and NF-κB protein in the colon; quantitative PCR to detect the relative expressions of TRAF6, IRAK1, NF-κB mRNA in the colon. ResultsCompared with the blank group, pathological changes of colon mucosa were severe in the model group, the contents of TNF-α, IL-1β and IL-6 increased remarkably, the expressions of TRAF6, IRAK1, p-NF-κB and NF-κB protein up-regulated in the colon (P<0.05). Compared with the model group, the contents of TNF-α, IL-1β and IL-6 decreased significantly in different dosages groups of Anchang decoction and mesalazine group (P<0.05); the expressions of TRAF6, IRAK1, p-NF-κB and NF-κB protein down-regulated in the colon (P<0.05). ConclusionAnchang decoction has therapeutic effects on TNBS-induced UC rats, and its effects might be related to that it could regulate TRAF6/IRAK1/NF-κB signaling pathway and the related inflammatory factors, thus relieving intestinal inflammatory reaction and intestinal mucosa injury.
