摘要:
目的运用网络药理学方法探讨柴胡配伍香附对癫痫靶基因及信号通路的影响。 方法通过中药系统药理学数据库与分析平台筛选柴胡、香附有效成分及靶点,然后采用人类基因数据库、在线孟德尔人类遗传数据库筛选与癫痫相关的基因,通过靶点交集获得中药与疾病的靶点,然后利用cytoscape.3.6.0软件绘制柴胡、香附有效成分与靶基因关系网络图,并对靶基因进行基因本体论功能富集分析、KEGG富集分析。 结果共筛选出柴胡119个、香附40个有效成分作用于癫痫的CHRM2、GABRA1、PRSS140等靶基因。靶基因主要富集在神经递质受体活性、类固醇激素受体、核受体活性、转录因子活性、乙酰胆碱受体活性等生物过程,涉及神经活性配体-受体相互作用、雌激素信号通路、胆碱能突触等信号通路。 结论从网络药理学角度筛选柴胡配伍香附治疗癫痫潜在的有效成分及作用靶基因和信号通路,可为开发柴胡配伍香附治疗癫痫提供理论依据。
Abstract:
ObjectiveTo discuss the effects of Chaihu (Bupleuri Radix) combined with Xiangfu (Cyperi rhizoma) on target gene and signaling pathway of epilepsy by using network pharmacology. MethodsThe active ingredients and targets of Chaihu and Xiangfu were screened from TCMSP, epilepsy-related genes were screened by adopting GeneCards and OMIM, the targets of TCM and diseases were obtained by the intersection of targets, and then network diagram of the relationship between active components and target genes of Chaihu and Xiangfu were drawn by utilizing cytoscape 3.6.0 software, GO and KEGG analysis of the target genes were carried out. ResultsAll 119 active ingredients are screened from Chaihu and 40 ones from Xiangfu, acting on epilepsy-related target genes including CHRM2, GABRA1 and PRSS140. Target genes are mainly enriched in biological processes such as neurotransmitter receptor activity, steroid hormone receptor, nuclear receptor activity, transcription factor activity and acetylcholine receptor activity, involving neuroactive ligand-receptor interaction, estrogen signal pathway, cholinergic synapse and other signal pathways. ConclusionThe potential active ingredients, the target genes and signaling pathway of Chaihu and Xiangfu in the treatment of epilepsy are selected from the perspective of network pharmacology, and it could provide theoretical basis for the development of Chaihu and Xiangfu in the treatment of epilepsy.
