摘要:
目的优化微乳超声提取葛根中总黄酮和总多糖的最佳条件,比较不同产地葛根微乳提取物对链脲佐菌素(streptozotocin,STZ)引起的2型糖尿病(type 2 diabetes mellitus,T2DM)小鼠的治疗作用。 方法以总黄酮和总多糖的提取率综合评分为指标,采用星点设计法优化微乳超声提取的最佳条件,通过STZ诱导建立T2DM小鼠模型,以二甲双胍为阳性对照药,对比湖南、陕西、湖北和山西地区葛根对T2DM小鼠的降血糖作用。记录各组小鼠第0、4、8、14天非空腹血糖值,第1、2、4、6、8、10、12、14天进食量,第0、2、4、6、8、10、12、14天体质量,并观察小鼠大小便和精神状态。 结果微乳提取的最佳条件为溶剂物料比:6.8倍,提取时间:68 min,提取功率:520 W;湖北地区葛根总黄酮和总多糖提取率最高,分别为4.40%和8.50%。湖北地区葛根微乳对T2DM小鼠的治疗效果最佳。 结论该微乳超声提取工艺稳定、操作简便、提取率高,且湖北地区葛根微乳提取物所含总黄酮和总多糖高于其他3个产地,其对降低STZ致T2DM小鼠血糖的作用最显著。
Abstract:
ObjectiveTo optimize the optimal conditions for ultrasonic extraction of total flavonoids and total polysaccharides from Gegen puerariae lobatae radix by microemulsion ultrasound, and compare therapeutic effects of microemulsion extract of Gegen from different habitats on STZ-induced T2DM mice. MethodsBy taking comprehensive scores of yield rate for total flavonoids and total polysaccharides as the indexes, central composite design was applied to optimize the best conditions of microemulsion ultrasounic extraction, T2DM mice models were established by STZ induction, to compare hypoglycemic activity of Gegen from Hunan, Shaanxi, Hubei and Shanxi on T2DM mice by selecting melbine as positive control medicine. To record non-fast blood glucose values on the zero, forth, eighth and fourteenth day, food intakes on the first, second, forth, sixth, eighth, tenth, twelfth and fourteenth day, body mass on the zero, second, forth, sixth, eighth, tenth, twelfth and fourteenth day of the mice in all the groups, to observe the urination, defecation and mental states of the mice. ResultsThe optimal conditions for microemulsion extraction is solvent to material ratio:6.8 times, extraction time was 68 min, extraction power: 520 W; the extraction percentages of total flavonoids and total polysaccharides from Gegen of Hubei were the highest, 4.40% and 8.50% respectively. Microemulsion of Gegen of Hubei presents the best therapeutic effects on T2DM mice. ConclusionThe microemulsion ultrasounic extraction process is stable, and easy to operate, showing high extraction rate, the contents of total flavonoids and total polysaccharides from Gegen of Hubei are higher than these of other three regions, and they present the most remarkable effects on reducing blood glucose of T2DM mice induced by STZ.
