摘要:
目的建立倒卵叶五加根与茎中总黄酮的含量测定方法,比较不同产地倒卵叶五加根、茎中总黄酮含量。 方法单因素试验筛选供试品制备方法,以硝酸铝-亚硝酸钠-氢氧化钠比色法测定总黄酮含量。 结果倒卵叶五加根与茎中总黄酮在0.162~0.975 mg范围内与吸光度呈良好线性关系,y=0.5318x-0.01,R2=0.9993,平均回收率为96.81%。根中总黄酮含量分别为陕西黄龙县(13.78±3.53)mg/g、陕西旬邑县(12.88±2.14)mg/g、陕西宜君县(8.78±4.22)mg/g、甘肃正宁县(9.03±3.81)mg/g(n=5);茎中总黄酮含量分别为陕西黄龙县(12.58±2.70)mg/g、陕西旬邑县(16.69±2.05)mg/g、陕西宜君县(10.28±2.87)mg/g、甘肃正宁县(14.09±3.49)mg/g(n=5)。 结论4个不同产地的倒卵叶五加药材根中总黄酮含量以陕西黄龙县最高,茎中总黄酮含量以陕西旬邑县最高;茎中总黄酮含量整体高于根中总黄酮含量。
Abstract:
ObjectiveTo establish the content determination methods of total flavonoids in the root and stem of Daoluanye Wujia (Acanthopanax obovatus Hoo), and compare the contents of total flavonoids of the plant from different producing areas. MethodsSingle factor test was used to screen the preparation method of the test sample and the content of total flavonoids was determined by aluminum nitrate-sodium nitrite-sodium hydroxide colorimetry. ResultsTotal flavonoids in the root and stem of Daoluanye Wujia showed good linear relationship in the range between 0.162 and 0.975 mg with the absorbance, y=0.5318x-0.01, R2=0.9993, average recovery rate was 96.81%. The contents of total flavonoids in the root ranked in the order: Shaanxi province Huanglong county (13.78±3.53)mg/g, Shaanxi province Xunyi county (12.88±2.14)mg/g, Shaanxi province Yijun county (8.78±4.22)mg/g, and Gansu province Zhengning county (9.03±3.81)mg/g (n=5); The contents of total flavonoids in the stem ranked in the order: Shaanxi province Huanglong county (12.58±2.70)mg/g, Shaanxi province Xunyi county (16.69±2.05)mg/g, Shaanxi province Yijun county (10.28±2.87)mg/g, and Gansu province Zhengning county (14.09±3.49)mg/g (n=5). ConclusionAmong four producing areas, the contents of total flavonoids are the highest in the root of Daoluanye Wujia from Shaanxi province Huanglong county, the contents of total flavonoids are the highest in the stem of the herb from Shaanxi province Xunyi county; the contents of total flavonoids in the stem are higher than these in the root generally.
