摘要:
目的考察五味子甲素对三硝基苯磺酸所致炎性肠病小鼠核转录因子κB(nuclear factor-κB,NF-κB)/环氧合酶2(cyclooxygenase,COX-2)通路的抑制作用。 方法采用三硝基苯磺酸灌肠诱导小鼠炎性肠病,采用ELISA、免疫组化、WB法测定小鼠体内NF-κB及COX-2表达,采用RT-PCR法测定小鼠结肠组织NF-κB及COX-2 mRNA表达,观察小鼠体质量、腹泻症状和疾病活动指数。 结果炎性肠病小鼠体内NF-κB、COX-2含量、蛋白及其mRNA表达增多,五味子甲素可抑制NF-κB、COX-2 mRNA和蛋白表达,抑制炎性肠病小鼠体质量减轻和疾病活动指数,改善小鼠腹泻情况。 结论五味子甲素可通过抑制NF-κB/COX-2通路的激活而抑制炎症反应,对三硝基苯磺酸诱导的炎性肠病小鼠具有治疗作用。
Abstract:
ObjectiveTo survey the inhibitory effects of deoxyschizandrin on NF-κB/COX-2 signal pathway in mice suffering TNBS-induced inflammatory bowel disease (IBD). MethodsTNBS enema was adopted to induce IBD in mice, ELISA, immunohistochemical method and WB method were applied to detect the expressions of NF-κB and COX-2 in mice, RT-PCR method was utilized to measure the expression of NF-κB and COX-2 mRNA, to observe body weight, the symptoms of diarrhea and disease activity index (DAI). ResultsThe expressions of NF-κB and COX-2 mRNA and protein increased in IBD mice, while deoxyschizandrin could inhibit the expressions of NF-κB and COX-2 mRNA and protein, restrain the reduction of body weight and DAI in IBD mice and improve diarrhea. ConclusionDeoxyschizandrin could inhibit inflammatory reaction through the inhibition of the activation of NF-κB/COX-2 signal pathway, thus showing therapeutic effects on TNBS-induced IBD in mice.
