摘要:
目的探讨蛇床子素对肝癌细胞自噬的影响。 方法通过体外实验,应用四甲基偶氮唑盐(methyl thiazolyl tetrazdium,MTT)法观察蛇床子素对Hepal-6小鼠肝癌细胞增殖抑制的影响,单丹磺酰尸胺(dansylcadaverine,MDC)染色法观察肝癌细胞自噬情况,免疫印迹法(Western blotting,WB)观察肝癌细胞自噬蛋白LC3和Beclin-1的表达情况。 结果MTT结果显示,蛇床子素能时间、剂量依赖性抑制Hepal-6小鼠肝癌细胞增殖(P<0.05);MDC染色结果显示,蛇床子素能促进Hepa1-6细胞自噬体增加;WB结果显示,蛇床子素能增加LC3-Ⅱ/LC3-I的表达比值,上调Beclin-1表达(P<0.05)。 结论蛇床子素能抑制Hepa1-6小鼠肝癌细胞增殖,促进其自噬体聚集,调控自噬蛋白表达。
Abstract:
ObjectiveTo discuss the influence of osthole on cellular autophagy of hepatoma carcinoma cell (HCC). MethodsMTT method was applied to observe the inhibitory effects of osthole on the proliferation of murine HCC Hepa1-6 via in-vitro experiments, MDC staining was used observe the autophagy of HCC, and WB to survey the expressions of HCC autophagy proteins LC3 and Beclin-1. ResultsMTT results showed that osthole could inhibit the proliferation of murine HCC Hepa1-6 in time-dependent and dose-dependent manner (P<0.05); MDC staining displayed that osthole could add autophagosome in Hepa1-6 cells; WB results revealed that the medicine could lift the ratio of LC3-Ⅱ/LC3-I, and upregulate the expressions of Beclin-1 (P<0.05). ConclusionOsthole could inhibit the proliferation of murine hepatoma carcinoma cells hepa1-6, promote the accumulation of autophagosomes and regulate the expression of autophagy proteins.
