摘要:
目的结合数据挖掘和网络药理学探究国家专利中药复方治疗黄疸型肝炎(icteric hepatitis,IH)的用药规律及作用机制。 方法首先在国家知识产权局中收集治疗IH的中药复方,利用中医传承计算平台V3.0软件和SPSS Modeler 18.0软件筛选出核心药对,然后运用TCMSP、PubChem、GeneCards、STRING、Metascape等平台检索核心药对的关键成分、靶点及作用机制并进行分析。 结果数据挖掘共获得133首国家专利中药复方,共434味中药;药性以寒性为多;药味以苦、甘为主;主要归肝、脾、肺经;功效以清热药、利水渗湿药居多;核心药对为茵陈-大黄。网络药理学共获得核心药对有效成分28种,药物靶点474个,IH相关作用靶点815个,交集靶点78个,关键成分有β-谷甾醇等,关键靶点有ALB、TP53、VEGFA等。GO富集分析结果显示,可能与免疫炎症反应、氧化应激反应、细胞凋亡等有关;KEGG通路富集分析结果显示,主要涉及雌激素、PI3K-AKT、MAPK、EB病毒感染等信号通路。 结论国家专利中药复方治疗IH的核心药对是茵陈-大黄,该核心药对通过多成分、多靶点、多途径发挥对IH的治疗作用。
Abstract:
ObjectiveTo survey the medication rules and the mechanism of national patent herbal compounds in the treatment of icteric hepatitis (IH) based on data mining and network pharmacology. MethodsHerbal compounds of treating IH were collected by China national intellectual property administration, to screen the core drug combinations using Traditional Chinese Medicine Inheritance Support System (TCMISS) V3.0 software and SPSS Moldeler 18.0 software, and then, TCMSP, PubChem, GeneCards, STRING and Metascape were applied to retrieve the key ingredients, the targets and the mechanism of the core couplet medicines. ResultsData mining obtained 133 national patented herbal compounds, involving 434 herbs, mostly of cold nature, mainly bitter and sweet flavors, entering the meridians of liver, spleen and lung, and belonging to the medicines of heat-clearing, clearing dampness and promoting-diuresis, and the core drug pairs were Yinchen (Artemisiae scopariae herba)-Dahuang (Rhei Radix et Rhizoma). The results of network pharmacology revealed that there were 28 active ingredients of the core herbal pairs, 474 herbal targets and 815 IH-related targets, as well as 78 intersected targets, and key ingredients containing β-sitosterin, and key targets ALB, TP53 and VEGFA. GO enrichment analysis displayed that it may be related to immune inflammation, oxidative stress, cellular apoptosis and others; KEGG pathway enrichment analysis showed that it mainly involved estrogen signaling pathway, PI3K-Akt, MAPK and EB virus infection. ConclusionThe core herbal pair for the treatment of IH with national patent herbal compounds contain Yinchen-Dahuang, and it could exert therapeutic effects on IH by many ingredients, many targets and many pathways.
