摘要:
目的基于网络药理学方法探讨大黄治疗糖尿病肾病(diabetic kidney disease,DKD)的潜在靶点与作用机制。 方法应用TCMSP、SwissTargetPrediction数据库筛选出大黄有效成分与相应靶蛋白;通过检索Drugbank、Disgenet数据库获得DKD的疾病靶点。采用Cytoscape 3.8.0构建大黄治疗DKD的“药物-活性成分-疾病-靶点”网络并进行拓扑结构分析;利用STRING平台进行蛋白互作网络分析;通过Metascape数据库进行基因本体(gene ontology,GO)和京都基因与基因组百科全书(kyoto encyclopedia of genes and genomes,KEGG)富集分析。 结果大黄治疗DKD的相关活性成分共7个,潜在作用靶点共55个。网络拓扑特征评价提示大黄可能通过作用于AKT1、TNF、TP53、CASP3、MTOR、IL-1β、PPARG、JUN、PTGS2及ESR1共10个核心靶点及相关通路达到治疗DKD的目的。 结论大黄具有多成分、多途径、多靶点协同作用的特点,主要通过干预氧化应激、细胞凋亡、炎症刺激及免疫反应等生物学过程改善或治疗DKD。
Abstract:
ObjectiveTo investigate the potential targets and mechanism of rhubarb (rhei radix et rhizoma) in the treatment of diabetic kidney disease (DKD) on the foundation of network pharmacology. MethodsThe active ingredients and the corresponding target protein of rhubarb were screened using TCMSP and SwissTarget-Prediction database; DKD - related targets were gained through searching Drugbank and Disgenet database. Cytoscape3.8.0 was applied to construct "medicine-active ingredients-disease-targets" network of rhubarb in the treatment of DKD, and to analyze topology structure; STRING platform was utilized to analyze PPI; Metascape database was applied to conduct GO and KEGG enrichment analysis. ResultsSeven related active ingredients of Rhubarb in treating DKD were obtained, involving 55 potential targets of action. The evaluation of network topology characteristics suggested that rhubarb could treat DKD possibly through acting on ten core targets and the related pathways such as AKT1, TNF, TP53, CASP3, MTOR, IL-1β, PPARG, JUN, PTGS2 and ESR1. ConclusionRhubarb could improve or treat DKD mainly by the intervention on the biological processes of oxidative stress, cellular apoptosis, inflammatory stimulation and immune response via the coordination of multiple ingredients, multiple pathways and multiple targets.
