摘要:
目的探讨推拿对坐骨神经损伤（chronic constriction injury，CCI）模型大鼠疼痛行为学、血清炎性介质及腓肠肌的影响。 方法将32只雄性SD大鼠随机分为4组，模型组及推拿组通过羊肠线轻扎大鼠坐骨神经方法制备CCI模型。空白组不予任何手术操作；假手术组不予羊肠线结扎，其余流程同CCI造模。干预后观察4组大鼠术侧行为学［机械足反射阈值（paw withdrawal threshold，PWT）、热缩足反射潜伏期（paw withdrawal latency，PWL）］、腓肠肌湿重恢复率、肌细胞形态学及血清白细胞介素1β（interleukin-1β，IL-1β）、IL-6、肿瘤坏死因子α（tumor necrosis factor-α，TNF-α）和5-羟色胺（5-hydroxytryptamine，5-HT）表达水平。 结果与空白组比较，模型组大鼠各时间点术侧PWT、PWL、腓肠肌湿重恢复率和肌细胞截面积均降低（P＜0.001），血浆IL-1β、IL-6、TNF-α和5-HT均升高（P＜0.001）；假手术组各指标与空白组相比均无统计学意义（P＞0.05）；与模型组比较，推拿组大鼠术侧PWT、PWL、腓肠肌湿重恢复率和肌细胞截面积均升高（P＜0.05），血浆IL-1β、IL-6、TNF-α和5-HT均降低（P＜0.05）。 结论推拿手法对CCI模型大鼠具有镇痛作用，其机制可能与抑制炎性介质IL-1β、IL-6、TNF-α和5-HT释放，减轻外周炎性反应有关。
Abstract:
ObjectiveTo explore the effects of massage on pain behavior, plasma inflammatory mediators and gastrocnemius muscle in chronic constriction injury (CCI) rat models. MethodsA total of 32 male SD rats were randomized into four groups, the model group and the massage group were prepared into CCI models through the method of light puncture of rat sciatic nerve with catgut. The blank group didn’t accept any surgical procedure, and sham operation group was not ligated with catgut, and the rest procedures were the same as CCI modeling. After the intervention, to observe surgical behavior including PWT and PWL, the recovery rate of gastrocnemius muscle wet weight, myocyte morphology, and the expressions of IL-1β, IL-6 and TNF-α as well as 5-HT in the four groups. ResultsCompared with the blank group, PWT and PWL, the recovery rate of gastrocnemius muscle wet weight, sectional area of myocytes in the model group were reduced at different time points (P<0.001), the levels of plasma IL-1β, IL-6 and TNF-α as well as 5-HT were increased (P<0.001); The difference had no statistical meaning when the indexes of sham operation group were compared with these of the blank group (P>0.05); compared with the model group, PWT and PWL, the recovery rate of gastrocnemius muscle wet weight, sectional area of myocytes were lifted in the massage group (P<0.05), the levels of plasma IL-1β, IL-6 and TNF-α as well as 5-HT were decreased (P<0.05). ConclusionMassage could relieve the pain in CCI rat models, and its mechanism might be related to restraining the release of inflammatory mediators including IL-1β, IL-6 and TNF-α, 5-HT, and alleviating peripheral inflammatory response.
