摘要:
目的观察七叶皂苷钠硬膜外腔注射对腰椎间盘突出症(lumbar disc herniation,LDH)模型大鼠背根神经节(dorsal root ganglion,DRG)神经元细胞电压门控钠离子通道(voltage-gated sodium channels,VGSCs)及相关炎性因子表达的影响。 方法将32只雄性SD大鼠随机分为空白组、假手术组、髓核组、七叶皂苷钠组,每组8只。造模成功后开始干预,七叶皂苷钠组以0.065 mg/kg剂量干预,其他3组不予药物干预,自由饮食水。干预后采用酶联免疫吸附试验(enzyme linked immunosorbent assay,ELISA)检测各组大鼠DRG组织肿瘤坏死因子α(tumor necrosis factor α,TNF-α)及白细胞介素6(interleukin-6,IL-6)表达水平;逆转录聚合酶链式反应(reverse transcription-PCR,RT-PCR)检测各组大鼠DRG组织中Nav1.3、Nav1.7、Nav1.8 mRMA表达;苏木精-伊红染色法(hematoxylin-eosin staining,HE)检测各组大鼠DRG组织形态学变化情况,并比较造模前1天及造模后第1、5、10、14天各组大鼠机械痛阀值和热痛阈值。 结果与髓核组比较,七叶皂苷钠组大鼠DRG组织形态学结构改善明显,机械痛阈值及热痛阀值升高(P<0.05),且DRG组织中Nav1.3、Nav1.7、Nav1.8 mRNA及TNF-α、IL-6表达降低(P<0.05)。 结论七叶皂苷钠硬膜外腔注射对LDH模型大鼠具有一定镇痛作用,其作用机制可能与七叶皂苷钠抑制Nav1.3、Nav1.7、Nav1.8及TNF-α、IL-6的表达有关。
Abstract:
ObjectiveTo observe the effects of epidural injection of sodium aescinate on the expressions of voltage-gated sodium channels (VGSCs) and related inflammatory factors in the neuronal cells of dorsal root ganglion (DRG) of the rats with lumbar disc herniation (LDH). MethodsAll 32 male SD rats were randomized into the blank group, sham operation group, nucleus pulposus group and sodium aescinate group, eight rats in each group. The intervenstion started after successfully modeling, sodium aescinate group was intervened at a dose of 0.065 mg/kg, other three groups remainded unhandled and they were on a free diet. After the intervention, ELISA was used to detect the expressions of TNF- α and IL-6 in DRG of the rats in different groups, RT-PCR was applied to detect the expressions of Nav1.3, Nav1.7 and Nav1.8 mRNA in DRG of the rats in different groups; and HE was adopted to observe histomorphological changes of DRG of the rats, to compare paw withdrawal threshold (PWT) and thermal pain thresholds of rats in each group on the day before modeling, the first, fifth, tenth and fourteenth day after modeling. ResultsCompared with nucleus pulposus group, the improvements of histomorphological structure of DRG were evident in sodium aescinate group, PWT and thermal pain thresholds were elevated (P<0.05), and the expressions of Nav1.3, Nav1.7 and Nav1.8mRNA, and the levels of TNF-α and IL-6 were reduced in DRG tissue (P<0.05). ConclusionEpidural injection of sodium aescinate has certain analgesic effects on LDH model rats, and its mechanism may be related to the inhibition of the levels of Nav1.3, Nav1.7 and Nav1.8, TNF-α and IL-6 by the medicine.
