摘要:
目的挖掘叶天士治疗痢疾的用药规律,探索其治疗痢疾核心药对的主要活性成分,初步研究治痢疾组方的治疗机制。 方法收集叶天士书籍中治疗痢疾的中药处方,采用中医传承辅助平台(V2.5)建立数据库,通过改进的互信息法、复杂系统熵聚类、无监督的熵层次聚类等方法,分析其用药规律,利用TCMSP、STRING Cytoscape软件等分析药对疾病间相互作用,对核心药物组合进行基因本体(gene ontology,GO)和京都基因与基因组百科全书(Kyoto encyclopedia of genes and genomes,KEGG)富集分析。 结果对筛选出的121例处方进行数据分析,确定处方中药物的四气五味、归经频次、用药频次,演化得到核心组合24个,关联规则分析得到置信度最高的药对为“黄芩-黄连”,共检索出192个靶点基因,痢疾相关疾病共检索出5836个靶基因,将两者相关靶基因进行匹配,得到共同靶基因163个,通过Cytoscape软件得出蛋白激酶B1(protein kinase B1,AKT1)、白细胞介素6(interleukin 6,IL-6)、血管内皮生长因子A(vascular endothelial growth factor A,VEGFA)、半胱氨酸天冬氨酸蛋白酶3(Caspase-3,CASP3)等靶点在黄芩-黄连治疗痢疾中具有重要地位和意义;GO功能富集分析得166条,主要涉及转录因子活性、酶活性、受体活性等生物功能,KEGG通路富集分析共有166个条目,主要调节人类巨细胞和卡波西氏肉瘤相关疱疹病毒感染、炎症、癌症等相关通路。 结论从中医传承平台角度分析,叶天士治疗痢疾以辨寒热虚实,攻补兼施,寒热并用,清热利湿,调气和血化滞为用药总则,用药灵活多变,种类丰富。通过网络药理学分析,叶天士用药具有多成分、多靶点和多途径的特点,核心药对主要通过调节巨细胞病毒(cytomegalovirus,CMV)感染、IL-17、肿瘤坏死因子(tumor necrosis factor,TNF)等免疫相关通路发生作用,为进一步治疗痢疾相对应疾病提供了新的思路,为深入研究方药作用机制奠定了基础。
Abstract:
ObjectiveTo primarily investigate the therapeutic mechanisms underlying prescriptions used in the treatment of dysentery through unearthing Ye Tianshi’s medication rule for the disease, and exploring the main active ingredients of the core drug combinations contained in the prescription. MethodsTCMISS was adopted to establish the data base after collecting herbal prescriptions for dysentery in Ye Tianshi’s book, the medication rule was analyzed through improved mutual information method, complex system entropy clustering, and unsupervised entropy hierarchical clustering, TCMSP and STRING Cytoscape were utilized to analyze drug-to-disease interactions, and to perform GO and KEGG enrichment analysis of the core drug combinations. ResultsThe study conducted data analysis of 121 selected prescriptions, confirmed four Qi and five flavors, the frequency of meridian tropism, medication frequency of the herbs in the prescriptions, and yielded 24 drug combinations. Association rule analysis identified "Huangqin (scutellariae radix)-Huanglian (coptidis rhizoma)" as the drug combination with the highest confidence, a total of 192 target genes were identified, while 5,836 target genes were found in dysentery-related diseases, 163 common target genes were obtained through pairwise matching, the targets including AKT1, IL-6, VEGFA and CASP3 played an important role in the treatment of dysentery with the drug combination via Cytoscape; GO enrichment analysis yielded 166 entries, mainly involving the biological functions such as the activity of transcription factor, enzyme and receptor, KEGG pathway enrichment analysis contained 166 entries, principally regulating the pathways of human cytomegalovirus and Kaposi’s sarcoma-associated herpesvirus infection, inflammation and cancer. Conclusion Ye Tianshi’s treatment for dysentery adopts differentiating cold and heat, deficiency and excess, tonification and purgation in combination, using cold and heat together, clearing heat and resolving dampness, regulating Qi, harmonizing blood and removing the stagnation as the general principles of medication via TCMISS. Ye Tianshi’s medication has the characteristics of multi-component, multi-target and multi-pathway through network pharmacology analysis, and the core drug combina-tions exert the function by adjusting CMV infection, IL-17, TNF and other immune-related pathways, and it could provide the new idea for further treatment of the corresponding diseases of dysentery, which has laid the foundation for in-depth study on the mechanism of the prescriptions and medicines.
