摘要:
目的采用网络药理学方法探索青黛治疗溃疡性结肠炎(ulcerative colitis,UC)的作用机制。 方法利用中药系统药理学数据库与分析平台筛选出青黛的有效成分及作用靶点;利用GeneCards数据库检索UC的相关基因,绘制药物靶点与疾病基因的韦恩图;利用Cytoscape构建化合物-疾病靶点的调控网络图;利用STRING数据库构建蛋白质-蛋白质相互作用网络(PPI);利用David 6.8软件将有效靶点进行基因本体论功能富集分析(gene ontology,GO)和KEGG富集分析(kyoto encyclopedia of genes and genomes,KEGG)。 结果青黛的活性成分有9个,主要有靛玉红、靛蓝和β-谷甾醇等;作用靶点100个,主要有转录因子AP-1、人前列腺素G/H合成酶2、半胱天冬酶3等;GO富集分析条目共84条,包括生物过程、细胞组成和分子功能;KEGG富集通路分析共93条通路,主要有细胞凋亡和炎症反应等多条信号通路。 结论青黛可能通过多靶点、多通路特性发挥治疗UC的作用。
Abstract:
ObjectiveTo explore the mechanism of Qingdai(Indigo Naturalis) in the treatment of ulcerative colitis (UC) based on network pharmacology. MethodsTCMSP was used to screen active ingredients and targets of Qingdai, UC-related genes were searched from GeneCards to draw Venn diagram of herbal targets and disease genes; Cytoscape was applied to construct regulatory network diagram of compound-disease targets; STRING database was utilized to build the networks of protein-protein interactions(PPI); and David 6.8 software to perform GO analysis and KEGG of the effective targets. ResultsThere were nine active ingredients in Qingdai, mainly containing Indirubin, indigo and β-sitosterol; and 100 targets, including JUN, PTGS2, CASP3; 84 items of GO enrichment analysis, covering biological process, cellular component and molecular function; and 93 pathways of KEGG enrichment analysis, embodying cellular apoptosis, inflammatory reaction and other signaling pathways. ConclusionQingdai could treat UC possibly through many targets and pathways.
