摘要:
目的运用数据挖掘分析鸡矢藤在脾胃病治疗的配伍规律，并用网络药理学探索作用机制。 方法收集处方信息建立数据库，用中医传承计算平台（the traditional Chinese medicine inheritance computing platform system，TCMICS）对药物进行统计分析，得到鸡矢藤核心组方后，利用中药系统药理学数据库与分析平台（traditional Chinese medicine systems pharmacology database and analysis platform，TCMSP）查找核心组方的有效成分及靶点；GeneCards和OMIM查找疾病基因，预测靶点；取得交集的靶点提交至Metascape数据平台进行基因功能注释及富集通路分析。 结果收集处方251张，共计98味药，化湿药总频次最多，鸡矢藤与姜半夏配伍频次最高；高频症状有胃胀、口干、口苦等；通过聚类算法得到核心组方：鸡矢藤、砂仁、姜半夏、炒麦芽和白术；网络药理学分析该核心组方通过槲皮素、山柰酚、黄芩素等有效成分，调控TP53、BCL2、RELA等靶点及MAPK信号通路、NF-κB信号通路、PI3K-Akt信号通路等达到调治脾胃病的目的。 结论通过数据分析归纳鸡矢藤在治疗脾胃相关疾病的用药规律，依托网络药理学分析鸡矢藤核心组方治疗脾胃病的作用机制，为后续研究和临床应用提供参考。
Abstract:
ObjectiveTo analyze the compatibility laws of Jishiteng in the treatment of spleen and stomach disease by using data mining, and explore the mechanism using network pharmacology. MethodsThe informa-tion of the prescription was collected to establish the database, TCMICS was used to perform statistical analysis of the herbs, in order to obtain the core prescription of Jishiteng, TCMSP was utilized to survey the active ingredients and the targets of the core prescriptions; GeneCards and OMIM to investigate the genes of disease and predict the targets; the targets of the intersection were obtained and submitted into Metascape data platform, to carry out the annotation of gene function and the analysis of enrichment pathway. ResultsAll 251 prescriptions were collected, involving 98 herbs, total frequency of the drugs for resolving dampness was the most, the frequency of Jishiteng combining with Jiangbanxia (Pinelliae Rhizoma Praeparatum Cum zingibere et alumine) was the highest; high-frequency symptoms contained gastric distension, dry mouth and bitter taste; the core prescriptions obtained by clustering algorithm were Jishiteng, Sharen (Amomi Fructus), Jiangbanxia, fried Maiya (Hordei fructus germinatus) and Baizhu (Atractylodis macrocephalae rhizoma); the analysis of network pharmacology indicated that the core prescriptions could regulate the targets including TP53, BCL2, RELA, and MAPK signaling pathway, NF-κB signaling pathway, PI3K-Akt signaling pathway through the active ingredients such as quercetin, kaempferol, and baicalein, so as to achieve the aim of regulating spleen and stomach. ConclusionMedication laws of Jishiteng in the treatment of spleen and stomach diseases concluded by data analysis, and the mechanism of the core prescrip-tion of Jishiteng in the treatment of spleen and stomach disease analyzed by network pharmacology could produce the reference for the subsequent study and clinical application.
