摘要:
目的:测定麻疼丸对糖尿病模型小鼠的镇痛作用及其改善微循环的作用,初步探讨麻疼丸治疗糖尿病周围神经病变的机理。方法:将72只昆明种小鼠随机分为空白对照组,模型对照组,阳性对照组及麻疼丸高、中、低剂量组,每组12只,除空白对照组外,其余各组采用四氧嘧啶造成糖尿病模型,按20mL/kg体质量容积灌胃给药,于72小时及20天后检测各组小鼠血糖浓度,用冷热痛觉测试仪测定热刺激时小鼠舔后足 时间和反应温度,及冷刺激时小鼠抱前爪时间和反应温度,用恒温水浴测定各小鼠甩尾时间;用显微镜测微尺测量各小鼠耳廓主动脉口径。结果:麻疼丸可不同程度地升高糖尿病模型小鼠尾痛阈和冷、热痛阈;麻疼丸对糖尿病模型小鼠耳壳血管口径有扩大作用,小鼠耳廓动脉微血管交叉数量明显增多。结论:麻疼丸能显著提高糖尿病模型小鼠的尾痛阈和冷、热痛阈,明显改善小鼠动脉微血管的循环,为麻疼丸治疗糖尿病患者周围神经病变提供一定的药理学依据。
Abstract:
Objective: To explore the mechanism of MaTengWan in treating diabetic peripheral neuropathy(DPN) preliminarily through determining the abirritation of MaTengWan to mice with diabetes and its function of improving microcirculation. Method: Seventy-two mice were randomized into blank control group, model control group, positive control group, high, moderate and low dose groups of MaTengWan, 12 mice each group. Except the mice in blank control group, diabetic models in other groups were established with alloxan, lavage with at a dose of 20mL/kg, blood glucose concentration of the mice in different groups were detected in 72 hours and 20 days. Time of licking hind feet and reaction temperature when there was thermal stimulation, time of holding forepaws and reaction temperature when there was cold stimulation, were determined with cold and hot pain sensation tester,tail-swinging time was determined with the method of thermostatic water bath; caliber of pinnal aorta in mice were measured with microscope micrometer. Result: MaTengWan could improve pain threshold and cold & hot pain threshold in different degrees; it could enlarge the caliber of pinnal aorta in diabetic mice, which could increase microvascular cross of pinnal aorta. Conclusion: MaTengWan could raise tail pain threshold and cold & hot pain threshold of diabetic mice significantly, improve microvascular circulation in artery of mice, which could provide definite pharmacological evidence for MaTengWan in treating DPN.
