摘要:
目的:观察血栓通的成分三七总皂苷(PNS)中单体皂苷:人参皂苷Rg1、三七皂苷R1等对受损的人脐静脉内皮细胞(HUVECs)的保护作用。方法:将不同的三七总皂苷单体皂苷人参皂苷Rg1、人参皂苷Rb1、三七皂苷R1分别与6~10 mmol/L的血管紧张素Ⅱ(AngⅡ)共同作用于HUVECs24小时后,采用硝酸还原酶法和放射免疫法分析技术分别测定HUVECs上清液中一氧化氮(NO)和内皮素(ET-1)含量。结果:AngⅡ组孵育24小时后,内皮细胞与空白对照组比较(P=0.000),NO释放明显减少(P=0.000),ET-1明显增加(P=0.000)。三种单体可显著抑制AngⅡ促ET-1分泌(P<0.05),促进HUVECs释放NO(P<0.05)。但三种单体组间比较,对NO及ET-1释放作用无统计学意义(P<0.05)。结论:AngⅡ可直接引起HUVECs的损伤与功能失调,血栓通三七总皂苷能够抑制血管紧张素Ⅱ引起的体外培养内皮细胞损伤,可保护内皮细胞结构及功能的完整性。
Abstract:
Objective: To investigate the protective effects of monomer saponin (ginsenoside Rg1, notoginse-noside R1 and others) in Panax notoginseng saponins (PNS) extracted from XueShuanTong on injuriedd human umbilical vein endothelial cells (HUVECs). Method: Different groups of cultured HUVECs were incubated with Ang Ⅱ(10~6 mmol/L) and ginsenoside Rg1, ginsenoside Rb1 and notoginsenoside R1 for 24 hours, the contents of Nitric oxide(NO) and endothelin-1(ET-1) in the HUVECs cultured medium were detected with nitrate reductase method and radioimmunoassay respectively. Result: Compared with blank control group, the release of NO in endothelial cells was reduced obviously (P=0.000), ET-1 was increased remarkably (P=0.000) after incubated for 24 hours. Three kinds of monomers could significantly inhibit the secretion of ET-1 induced by AngⅡ(P<0.05) and promote HUVECs releasing NO(P<0.05), but the comparisons among three kinds of monomers showed no statistical meaning in NO and the release of ET-1 (P<0.05). Conclusion: AngⅡ could directly cause the injury of HUVECs and dysfunction, PNS extracted from XueShuangTong could inhibit endothelial injury induced by AngⅡ, protect the integrity of endothelial structure and function
