摘要:
目的:通过不同浓度白头翁水煎液对离体蛙心功能的影响及其机制的研究,为新药的研制和深入临床实践提供理论依据。方法:采用斯氏蛙心灌流法制备离体蛙心标本,分别加入不同浓度(100%,50%,25%,12.5%)的白头翁水煎液(0.1 mL),观察药物对离体蛙心的作用,并了解是否存在剂量依赖性。加入0.05 mL、0.1 mg/mL盐酸维拉帕米溶液,观察并记录实验数据,然后加入白头翁水煎液,观察药物对离体蛙心的作用;加入0.05 mL、1 mg/mL盐酸普罗帕酮溶液,观察并记录实验数据,然后加入白头翁水煎液,观察药物对离体蛙心的作用。探讨其作用机制与钠、钙通道的关系。结果:4种不同浓度的白头翁水煎液均能有效地增强蛙心的收缩力,使蛙心收缩频率减慢。同时白头翁水煎液能逆转盐酸普罗帕酮对心脏的影响,但不能逆转盐酸维拉帕米对心脏的作用。结论:白头翁水煎液对离体蛙心有明显的影响,能逆转盐酸普罗帕酮溶液对心脏的作用。
Abstract:
Objective: To provide theoretical basis for the development of new drug and clinical practice by investigating the effects of BaiTouWeng(radix pulsatilla) decoction in different concentrations on isolated frog heart and the mechanism. Methods: The specimens of isolated frog hearts were established by the method of frog heart perfusion, the effects of different concentrations of BaiTouWeng decoction (100%, 50%, 25% and 12.5%) on isolated frog hearts were observed respectively, to investigate whether the dose-dependent manner was existed. The specimens were administered with verapamil hydrochloride, 0.05 mL, 0.1 mg/mL, experimental data was recorded and observed, BaiTouWeng decoction was added to survey its effects on isolated frog heart; the specimens were given with propafenone hydrochloride, 0.05 mL, 0.1 mg/mL, to record and observe experimental data, BaiTouWeng decoction was added to survey its effects on isolated frog heart, the relationship among the mechanism, sodium and calcium channel was explored. Results: BaiTouWeng decoction with different concentrations could effectively enhance cardiac contractility of the frogs, at the same time slow down contraction frequency of frog heart. It could also reverse the effects of propafenone hydrochloride on frog heart, but showed no effects of verapamil hydrochloride influencing on the heart. Conclusion: BaiTouWeng decoction demonstrates obvious action on isolated frog heart and reverses the influence of propafenone hydrochloride on the heart.
