摘要:
目的:通过研究苦参碱对CCl4诱导的肝纤维化大鼠肝脏转化生长因子β1表达的影响,探讨苦参碱对肝纤维化的抑制作用。方法:CCl4诱导大鼠肝纤维化动物模型,免疫组化方法观察苦参碱对肝纤维化大鼠肝脏转化生长因子β1表达的变化,并分别检测血清学及肝脏组织学变化。结果:与肝纤维化模型组相比,苦参碱组大鼠血清谷草转氨酶、丙氨酸转氨酶水平明显降低(P<0.05),HE染色未见假小叶形成,在汇管区周围,免疫组化染色显示部分肝细胞胞浆及纤维间隔区及炎性细胞浸润区转化生长因子β1的表达明显减少(P<0.05)。结论:苦参碱可以抑制纤维化的肝组织转化生长因子β1的表达,明显改善肝纤维化大鼠的肝组织结构,抑制肝纤维化的进展。
Abstract:
Objective: To explore the inhibition of matrine to liver fibrosis by studying the effects of matrine on the expressions of transforming growth factor β1(TGF-β1) of liver in the rats with liver fibrosis induced by CC14. Methods: Animal model with liver fibrosis was induced with CC14, the effects of matrine on the expressions of TGF-β1 in rats with liver fibrosis were observed with immunohistochemical method, the changes of serology and liver histology were detected respectively. Results: Compared with the model group of liver fibrosis, the levels of serum glutamic oxalacetic transaminase (SGOT) and alanine aminotransferase of the rats in the matrine group were decreased notably (P<0.05), HE staining didn't show the formation of pesudolobuli, immunohistochemical staining demonstrated that the expressions of TGF-β1 became less in the cytoplasm of liver cells, fibrosis spacer and absorption field of the inflammatory cells around the portal area (P<0.05). Conclusion: Matrine could inhibit the expressions of TGF-β1 in the liver tissue, obviously improve the construction of liver tissue in rats with liver fibrosis, and inhibit the progression of liver fibrosis.
