摘要:
目的:观察黄鼬干粉胶囊对再生障碍性贫血(AA)模型小鼠造血功能的影响及作用机制。方法:采用X射线+环磷酰胺+氯霉素复合方法复制小鼠AA模型,观察黄鼬干粉胶囊对小鼠外周血象、骨髓有核细胞数(BMNC)、骨髓粒细胞-巨噬细胞集落生成单位(CFU-GM)、血清中可溶性细胞间黏附因子1(sICAM-1)和骨髓组织病理学的影响。结果:黄鼬干粉胶囊能明显增加模型小鼠外周血中HGB、RBC、WBC和PLT数量,能增加骨髓BMNC数量,还能升高骨髓CFU-GM和降低血清sICAM-1。与模型对照组比较,差异有统计学意义(P<0.05)。骨髓组织病理学结果表明,黄鼬干粉能明显改善模型小鼠骨髓增生低下和造血灶减少,促进骨髓造血。结论:黄鼬干粉胶囊能改善AA模型小鼠造血功能,其作用机制可能与调节骨髓CFU-GM和血清sICAM-1的平衡有关。
Abstract:
Objective: To observe the influence of yellow weasel dry powder capsules on hemopoiesis of mice model with aplastic anemia (AA) and the mechanism. Methods: Mice model with AA was prepared by X-ray, cyclophosphamide and chloramghenicol, the influence of yellow weasel dry powder capsules on peripheral blood, bone marrow mononuclear cells (BMNC), colony forming unit-granulocyte and macrophage (CFU-GM), soluble intercellular adhesion molecules-1(sICAM-1) and the histopathology of bone marrow were observed. Results: Yellow weasel dry powder capsules could increase the numbers of HGB, RBC, WBC and PLT in peripheral blood of the mice obviously, increase the amount of BMNC, raise CFU-GM and decrease sICAM-1. Compared with model control group, the difference showed statistical meaning (P<0.05). Histopathological results of bone marrow demonstrated that yellow weasel dry powder could notably improve hypofunction of bone marrow hyperplasia and the decrease of hemopoietic focus of AA mice, and promote hemopoiesis of bone marrow. Conclusion: Yellow weasel dry powder capsules could effectively improve hemopoiesis of AA mice model, and its mechanism might be related to regulating the balance of CFU-GM and sICAM-1.
