摘要:
目的:通过观察镰形棘豆总黄酮含药血清干预转化生长因子β1(TGF-β1)诱导的人肾小管上皮细胞(HK-2)纤维化细胞因子——血管内皮生长因子(VEGF)mRNA的表达,进一步探讨其在防治肾间质纤维化方面的作用。方法:将HK-2 细胞用含10%胎牛血清的DMEM/F12(1∶1)培养基培养;实验分为4 组:空白对照组、TGF-β1诱导组(TGF-β110 ng/mL)、空白血清对照组(TGF-β110 ng/mL +10%空白血清)、镰形棘豆总黄酮组(TGF-β110 ng/mL+10%镰形棘豆总黄酮含药血清)。药物干预24小时后,荧光定量PCR检测VEGF mRNA的表达。结果:HK-2细胞经TGF-β1诱导后,VEGF mRNA的表达显著上升,与空白对照组比较差异有统计学意义(P<0.05),经镰形棘豆总黄酮药物血清干预后,VEGF mRNA的表达逐步下降,与单纯TGF-β1诱导组比较差异有统计学意义(P<0.05)。结论:镰形棘豆总黄酮在一定程度上能够抑制TGF-β1诱导的人肾小管上皮细胞纤维化,其机制可能与调节纤维化细胞因子VEGF的mRNA表达有关。
Abstract:
Objective: To explore the effects of preventing and treating renal interstitial fibrosis of total flavonoids of oxytropis falcata further by observing the intervention effects on HK-2 and (VEGF) mRNA expressions induced by TGF-β1. Methods: HK-2 cell was cultivated by using DMEM/F12(1∶1) medium containing 10% fetal bovine serum; the experiments were divided into four groups: blank control group, TGF-β1 induced group (TGF-β1 10 ng/mL), blank serum control group (TGF-β110 ng/mL +10% blank serum), total flavonoids of oxytropis falcate(TGF-β110 ng/mL+10%drug serum of total flavonoids of oxytropis falcata ). After 24 hours of drug intervention, VEGF mRNA expression was detected by fluorescent quantitative PCR. Results: After HK-2 cell was induced by TGF-β1, VEGF mRNA expression increased obviously, there was statistical difference compared with blank control group(P<0.05), after intervention of drug serum of total flavonoids of oxytropis falcata, VEGF mRNA expression decreased gradually, there was statistical differences compared with simple TGF-β1 induced group(P<0.05). Conclusion: Total flavonoids of oxytropis falcate can inhibit human renal tubular epithelial cell fibrosis induced by TGF-β1 to a certain degree, and its mechanism might be related to fibrotic VEGF mRNA expression regulation.
