摘要:
目的:研究四物汤水提液对脑缺血再灌注小鼠海马组织形态的影响及脑组织SOD和GSH-PX活力的影响,进一步探讨四物汤水提液对脑缺血再灌注保护作用的机制。方法:将55只雄性健康昆明种小鼠,随机分成脑缺血再灌注模型对照组,假手术组,四物汤水提液高、中、低剂量组,每组11只。采用Himori法制备小鼠脑缺血再灌注模型,四物汤水提液高、中、低剂量组分别灌胃四物汤水提液(40、20、10 g/kg),连续给药15天,模型对照组及假手术组给予等体积蒸馏水。于末次给药后1小时,处死小鼠,取海马组织,制作病理标本,光镜下观察小鼠海马组织形态学变化。取小鼠脑组织,测定SOD和GSH-PX的活力。结果:与假手术组相比,模型组小鼠海马神经细胞结构明显异常;与模型组比较,四物汤水提液高、中、低剂量组小鼠海马神经细胞结构异常均有改善;与假手术组相比,模型组小鼠脑组织中SOD和GSH-PX的活力均显著降低(P<0.05),与模型组比较,四物汤水提液高、中、低剂量组小鼠脑组织中SOD和GSH-PX的活力均有显著提高(P<0.05)。结论:四物汤水提液对脑缺血再灌注小鼠海马组织病理改变具有一定的保护作用,其保护作用可能与其抗氧化作用有关。
Abstract:
Objective: To explore the effects of water extraction of SiWu decoction on hippocampus and activity of brain tissue SOD and GSH-PX in mice with cerebral ischemia-reperfusion, and to discuss protection mechanism of water extraction of SiWu decoction on cerebral ischemia-reperfusion furtherly. Methods: 55 healthy male mice from Kunming were randomly divided into the model control group, sham operation group, the low, moderate and high dosage groups of water extraction of SiWu decoction, each group eleven mice. mice models with cerebral ischemia-reperfusion were made by using Himori methods, the low, moderate and high dosage groups of waterextraction of SiWu decoction were given intragastric administration of water extraction of SiWu decoction(40,20,10 g/kg), continuous medication for 15 days, the model control group and the sham operation group were given isovolumetric distilled water. After one hour of the last medication, mice were killed, hippocampi were aquired, pathological samples were made, histomorphology changes of mice hippocampi were observed under light microscope. Cerebral tissue were fetched, then the activity of SOD and GSH-PX were measured. Results: Compared with the sham operation group, neuron structure of mice hippocampus in the model group was obvious abnormal; compared with the model group, the abnormal neuron structure of mice hippocampus in the low, moderate and high dosage groups of water extraction of SiWu decoction all improved; compared with the sham operation group, activity of SOD and GSH-PX of cerebral tissue in the model group decreased obviously(P>0.05), compared with the model group, activity of SOD and GSH-PX of cerebral tissue in the low, moderate and high dosage groups of water extraction of SiWu decoction increased significantly(P<0.05). Conclusion: Water extract of SiWu decoction has certain protection against of histomorphology changes of hippocampus mice with cerebral ischemia-reperfusion, its protection may has relations with antioxidant function.
