摘要:
目的:探讨阿里红多糖(FoP)对小鼠体外免疫的调节作用。方法:采用体外淋巴细胞增殖实验考察FoP对小鼠脾淋巴细胞增殖的影响;采用磺酰罗丹明B(SRB)法检测不同浓度FoP单独作用及其与有丝分裂原ConA、LPS共同作用对小鼠脾淋巴细胞增殖的影响;采用酶联免疫吸附实验(ELSA)法检测不同浓度FoP体外刺激对小鼠脾淋巴细胞诱生白介素2(IL-2)、白介素13(IL-13)、肿瘤坏死因子α(TNF-α)等细胞因子的影响。结果:Fop在25~100μg/mL浓度范围内能显著促进小鼠脾淋巴细胞增殖,并与ConA、LPS协同促进T、B淋巴细胞增殖,还能诱导脾淋巴细胞分泌IL-2、TNF-α。结论:FoP具有免疫促进作用。
Abstract:
Objective: To explore the immunoregulation of fomes oficinalis sporophore polysaccharide (FoP) on the mice in vitro. Methods: Lymphocyte proliferation test was adopted to investigate the effects of FoP on the proliferation of splenic lymphocyte of the mice; and SRB method was used to detect the act of FoP independently in different concentrations and its combined action with the mitogen ConA, LPS on splenic lymphocyte of the mice; ELISA method was adopted to survey the effects of FoP with different concentrations in vitro stimulating splenic lymphocyte inducing the cell factors such as IL-2, IL-13, TNF-α and others. Results: FoP could promote the proliferation of splenic lymphocyte of the mice in the concentrations ranged between 25 and 100 μg/mL, and promote T and B lymphocyte proliferation by working with ConA, LPS, and induce splenic lymphocyte secreting IL-2 and TNF-α. Conclusion: FoP plays a role in immunological enhancement.
