摘要:
目的:对陇中损伤散进行毒理学实验研究,对其安全性进行客观评价,为临床药物的研制与开发提供确实实验数据。方法:急性毒性实验以最大浓度、最大体积的陇中损伤散灌胃小鼠,测定1日最大给药量。长期毒性实验以高、中、低3个剂量的陇中损伤散连续灌胃大鼠90天,观察大鼠的生长发育、血液学、血液生化学、脏器系数、组织病理变化及停药后14天上述指标的变化。结果:急性毒性实验结果表明小鼠1日内的最大给药量为生药49.6 g/kg体质量,相当于临床成人每日用量0.05 g/kg体质量的992倍。长期毒性实验结果表明给药90天时,高剂量组丙氨酸氨基转移酶(ALT)较对照组升高(P<0.01),高剂量组肝重量系数增高(P<0.05)。停药14天后,大鼠行为、体质量、血液学、血液生化学、脏器系数、组织病理学检查均未见明显异常(P>0.05)。结论:按临床规定的疗程、剂量用药是安全、有效的。
Abstract:
Objective: To provide experimental data for the manufacture and development of clinical drugs by exploring the experimental study of toxicology in Longzhong SunShang powder, and evaluating the safety objectively. Methods: The mice were fed with Longzhong SunShang powder in the acute toxicity test with the maximum concentration and volume, and the maximum dose in one day was measured. The rats were fed with high, medium and low doses of Longzhong SunShang powder for 90 days in the long-term toxicity test, and the index changes of the growth and development, hematology, blood biochemistry, organ coefficient, pathological changes as well as 14 days after drug withdrawal of rats were observed. Results: The results of the acute toxicity test indicated that the maximum dose of the crude drug for the mice were 49.6 g/kg body weight, which was equivalent to 992 times of adult clinical daily dosage of 0.05 g/kg body weight. The long-term toxicity test showed that alanine aminotransferase (ALT) in the high dose group was higher than that in the control group at the ninetieth day (P < 0.01), and the liver weight coefficient in the high dose group increased (P <0.05). After 14 days' withdrawal, the behavior, weight, hematology, blood biochemistry, organ coefficient and histopathological examination of the rats showed no obvious abnormality (P>0.05). Conclusion: It is safe and effective when the medication follows the prescribed course of treatment and dosage.
