摘要:
目的:研究当归水煎液化学成分体内代谢情况,结合肠吸收遴选当归质量控制成分。方法:采用翻转肠囊法与腹主动脉采血试验,结合高效液相色谱法(HPLC)指纹图谱,研究当归水煎液化学成分的肠吸收与入血情况。结果:通过翻转肠囊实验,明确了22种通过肠吸收的成分,其中成分1、3、4、5、6、7、11、12、13、14、15、16(阿魏酸)、17、18(洋川芎内酯I)、19(洋川芎内酯H)、20、21、22以原型成分被肠吸收,成分M1、M2、M3、M4是通过肠壁酶代谢的新成分。分析腹主动脉采血HPLC明确了5种成分化合物12、18(洋川芎内酯I)、19(洋川芎内酯H)、M5、M6入血,其中成分12、18(洋川芎内酯I)、19(洋川芎内酯H)可直接通过肠吸收入血,成分M5、M6为经过代谢吸收入血的新成分。结论:综合肠吸收实验与入血成分分析,初步遴选出成分12、18(洋川芎内酯I)、19(洋川芎内酯H)作为当归的质量控制成分。
Abstract:
Objective: To study the metabolism in vivo of chemical composition in the decoction of DangGui (Angelica sinensis) and select the quality control component of DangGui by combining with intestinal absorption. Methods: The everted gut sac method and abdominal aorta blood sampling test as well as the combination of HPLC fingerprinting were adopted to investigate the absorption and blood supply of chemical constituents in the decoction of DangGui. Results: Twenty-two intestinal absorption components were identified by everted gut sac tests, and among which, components 1, 3, 4, 5, 6, 7, 11, 12, 13, 14, 15 and 16 (ferulaic acid), 17, 18 (Senkyunolide I), 19 (Senkyunolide H), 20, 21 and 22 were absorbed by intestine with prototype components. At the same time, M1, M2, M3 and M4 we are the new components with enzymatic metabolism through the intestinal wall. It was identified by the analysis of abdominal aortic blood HPLC that the five compounds 12, 18 (senkyunolide I), 19 (senkyunolide H), M5, M6 were in blood. Among which, the composition of 12, 18 (senkyunolide I) and 19 (senkyunolide H) were directly absorbed into the blood through intestine while M5 and M6 were the new components absorbed into the blood by metabolism. Conclusion: Components 12, 18 (senkyunolide I) and 19 (senkyunolide H) are selected preliminarily as the quality control components of DangGui by the analysis of intestinal absorption experiment and blood intake components.
