摘要:
目的:建立骆驼蓬子中去氢骆驼蓬碱对照品的制备分离方法。方法:通过AB-8大孔树脂柱色谱分离,重结晶,HPLC制备色谱分离纯化,从骆驼蓬子的70%乙醇提取物分离得到一个化合物,通过NMR、M S等波谱方法鉴定该化合物为去氢骆驼蓬碱,经TLC,HPLC-UV,HPLC-ELSD及MS联用等技术检验其纯度。结果:从骆驼蓬子中制备分离的去氢骆驼蓬碱对照品,质量分数>98%。结论:本法所得的去氢骆驼蓬碱对照品符合中药化学对照品的相关技术要求,为药材骆驼蓬子和含骆驼蓬子的成方制剂的质量控制及药效物质基础研究提供化学对照品。
Abstract:
Objective: To establish the preparat on separation method of the control article of harmine in LuoTuo PengZi (Peganum harmala L). Methods: Chromatography separation and purification was prepared by AB-8 macroporous resin column chromatography separation, recrystallization and HPLC. A compound was isolated from the 70% ethanol extract of LuoTuo PengZi and identified as harmine by NMR, MS and other spectroscopy methods. The purity was tested by the combination of TLC, HPLC-UV, HPLC-ELSD and MS. Results: The mass fraction of the control article of harmine isolated and prepared from LuoTuo PengZi was greater than 98%. Conclusion: The control article of harmine obtained by this method conforms to the relevant technical requirements standard of traditional Chinese medicine chemical reference substance. It could provide chemical reference substance for the quality control and material basis of pharmacodynamics of LuoTuo PengZi and patent medicine which contains LuoTuo PengZi.
