摘要:
目的:探讨山茱萸对老龄小鼠肝微粒体细胞色素P450部分亚型的影响。方法:对12月龄小鼠给予含山茱萸不同剂量的药物鼠粮,连续给药3个月后,以6月龄小鼠为成年对照组,采用荧光定量PCR法检测小鼠肝脏组织CYP1A1、CYP2A4、CYP2A5、CYP3A41等P450亚型基因mRNA的表达。结果:模型组小鼠肝脏组织CYP1A1、CYP2A4、CYP2A5、CYP3A41等基因mRNA的表达量显著低于正常组小鼠;不同剂量山茱萸给药组肝脏CYP1A1、CYP2A4、CYP2A5、CYP3A41等基因mRNA的表达量均显著高于模型组,且山茱萸高剂量组作用优于低剂量组。结论:衰老可导致细胞色素P450亚型基因表达减少,山茱萸对老龄小鼠肝微粒体CYP1A1、CYP2A4、CYP2A5、CYP3A41等基因具有诱导作用。
Abstract:
Objective: To observe the effects of fructus corni (Cornus officinalis Sieb. et Zucc.) on cytochrome (CY) P450 partial subtype of liver microsome of the aged mice. Methods: The mice aged 12 months were given food containing different doses of fructus corni for three months consecutively, the mice aged six months were taken as the control group, the expressions of CY P450 subtype gene mRNA including CYP1Al, CYP2A4, CYP2A5, CYP3A41 and others in liver tissue of the mice were detected by using fluorescent quantitative PCR method. Results: The expressions of genetic mRNA including CYP1Al, CYP2A4, CYP2A5, CYP3A41 and others in the liver tissue of the mice in the model group were lower than these of the mice in the normal group; the expressions of genetic mRNA including CYP1Al, CYP2A4, CYP2A5, CYP3A41 and others in the liver tissue of the mice in different doses groups of fructus corni were higher than these of the model group significantly, and high dose group was superior to low dose groups. Conclusion: Senility could cause the expression reduction of CY P450 subtype gene, while fructus corni could induce the genes of CYP1Al, CYP2A4, CYP2A5, CYP3A41 and others in liver microsome of the aged mice.
