摘要:
目的:探讨武权生教授周期疗法对乳腺增生病模型大鼠雌激素受体(ER)、孕激素受体(PR)、细胞外调节蛋白激酶(ERK)表达的影响。方法:将90只大鼠随机分为空白组、模型组、逍遥丸组、三苯氧胺组及中药低、中、高剂量组,除空白组外其余各组均进行乳腺增生造模。造模成功后空白组和模型组给予生理盐水0.2 mL/(kg·d)灌胃,其余5组分别给予逍遥丸、三苯氧胺、中药(Ⅰ号方及Ⅱ号方低、中、高剂量)灌胃,共30天。结果:造模后,各造模组大鼠的乳头直径与空白组比较差异有统计学意义(P<0.05)。灌服Ⅰ号方后各用药组大鼠乳头直径缩小,但仍大于空白组(P<0.05)。灌服Ⅱ号方后大鼠的乳头直径进一步缩小,且小于模型组(P<0.05),除中药高剂量组外其余各组乳头直径大于空白组(P<0.05);各组ER、PR、ERK积分低于模型组(P<0.05),中药高剂量组ERK积分低于三苯氧胺组(P<0.05)。结论:武权生应用中药周期疗法可使乳腺增生模型大鼠增生乳头缩小,ER、PR、ERK积分降低。
Abstract:
Objective: To explore the effects of professor Wu Quansheng′s periodic treatment empirical formula on ER, PR and ERK of the rat model with hyperplasia of mammary gland(HMG). Methods: Ninety rats were randomized into the blank group, the model group, XiaoYao pills group, tamoxifen group, low, moderate and high dosages groups of herbs, except the blank group, the models with HMG were set up in the other groups. After successfully establishing the models, the blank group and the model group were drenched with physiological saline, 0.2 mL/( kg·d), the other five groups were gavaged with XiaoYao pills, tamoxifen and herbs (prescription no.1 and prescription no.2 in low, moderate and high dose), for 30 days. Results: After establishing the models, the difference had statistical meaning in the comparisons of mammilla diameter of the rats in different model groups when they were compared with the blank group (P<0.05). Mammilla diameters of the rats reduced in all the medication groups after drenching with prescription no.1, but still larger than the blank group (P<0.05). Mammilla diameters of the rats reduced further after drenching with prescription no.2, while still smaller than the model group (P<0.05), except high dosage group of herbs, mammilla diameters of the rats in other groups were larger than these of the blank group (P<0.05); ER, PR and ERK scales of different groups were lower than these of the model group (P<0.05); high dosage group of herbs was lower than tamoxifen group in ERK scales (P<0.05). Conclusion: Wu Quansheng′s periodic treatment could reduce mammilla diameters and reduce ER, PR and ERK of the rat model with HMG.
