摘要:
目的:研究海藻4种不同方法提取物的体外抗病毒活性。方法:以呼吸道合胞病毒(RSV)、单纯疱疹病毒Ⅰ型(HSV-Ⅰ)、柯萨奇病毒B5(COXB5)、肠道病毒71型(EV71)4种病毒感染的人喉癌上皮细胞(Hep2)、横纹肌瘤细胞(RD)为模型,运用4种不同方法提取的海藻提取物对4种病毒感染的细胞进行治疗,并设利巴韦林、阿昔洛韦作为阳性对照药。采用细胞病变效应法(CPE法)结合4,5-二甲基噻唑-2,5-二苯基四唑溴化物(MTT)比色法检测细胞存活率,计算药物的治疗指数TI值,分析4种不同提取方法提取的海藻提取物对4种病毒的抑制作用。结果:海藻各提取物均有不同程度的抗病毒活性,其中水提取物抑制作用最强,其对RSV、HSV-Ⅰ、COXB5、EV71的治疗指数TI值分别为413.00、310.83、7.36、176.07,与阳性对照药效果相当;乙醇超声提取物对COXB5效果较好,TI值为19.97。结论:海藻提取物具有体外抗RSV、HSV-Ⅰ、COXB5、EV71活性的作用。
Abstract:
Objective To study in-vitro antiviral activity of seaweed extracts obtained by four different methods. Methods: Hep2 and RD cells were used to as the model and infected by four viruses including RSV, HSV-Ⅰ, COXB5 and EV71, and the infected cells were treated by seaweed extracts obtained by four methods, ribavirin and acyclovir were set as positive control drug. Cellular survival rate was detected by cytopathic effect(CPE) method and MTT colorimetry, therapeutic index TI value of the drugs was calculated, and the inhibitory actions of seaweed extracts obtained by four methods to four viruses were analyzed. Results: Seaweed extracts possessed antiviral activity in different degrees, especially the strongest inhibitory effects of water extract, and its TI values to RSV, HSV-Ⅰ, COXB5 and EV71 were 413.00, 310.83, 7.36 and 176.07 respectively, equivalent to the effects of positive control drugs. Ethanol ultrasonic extract showed the better effects to COXB5 and TI value was 19.97. Conclusion: Seaweed extracts possessed in-vitro antiviral activity to RSV, HSV-Ⅰ, COXB5 and EV71.
