摘要:
目的:观察富硒黄芪提取物对免疫抑制大鼠细胞因子白细胞介素2(IL-2)、白细胞介素(IL-4)、干扰素γ(IFN-γ)及肿瘤坏死因子α(TNF-α)的影响。方法:将SD大鼠随机分为6组,空白组、模型组及富硒黄芪提取物高、中、低剂量组和普通黄芪提取物组。除空白组外,其余5组均采用腹腔注射环磷酰胺构建大鼠免疫抑制模型。造模成功后分组干预治疗。给药第7天后,各组大鼠禁食8小时,股动脉取血,采用酶联免疫吸附法(ELISA)检测血清中IL-2、IL-4、IFN-γ及TNF-α的含量。取出胸腺、脾脏记录其重量变化。结果:与空白组比较,模型组大鼠胸腺、脾脏指数显著降低,差异有统计学意义(P<0.05);与模型组比较,各干预组大鼠胸腺、脾脏指数有所升高,差异有统计学意义(P<0.05)。与空白组比较,模型组大鼠细胞因子(IL-2、IL-4、IFN-γ)含量显著降低,TNF-α含量显著升高,差异有统计学意义(P<0.05);与模型组比较,各干预组大鼠IL-2、IL-4、IFN-γ含量有所升高,TNF-α含量有所降低,差异有统计学意义(P<0.05)。结论:富硒黄芪提取物能显著提高环磷酰胺所致免疫抑制模型大鼠的免疫功能,且这种作用较普通黄芪更为显著。
Abstract:
Objective: To observe the effects of extract of selenium-enriched Astragalus membranaceus on cytokines IL-2, IL-4, IFN-γ and TNF-α in immunosuppressive rats. Methods: SD rats were randomly divided into six groups: the blank group, the model group, the high, middle and low dose groups of extract of selenium-enriched Astragalus membranaceus as well as the common astragalus extract group. Other five groups were injected intraperitoneally with cyclophosphamide to construct the immunosuppressive models except the blank group. After the modeling, the groups were intervened respectively. After seven days of treatment, rats, fasted for eight hours, were blooded from the femoral artery. The contents of cytokines IL-2, IL-4, IFN-γ and TNF-α in the serum of immunosuppressive rats were detected by enzyme-linked immunosorbent assay ELISA, thymus gland and the spleen were taken out and the changes were recorded. Results: Compared with the blank group, thymus and spleen indexes in the model group decreased significantly, and the difference was statistically significant(P<0.05); Compared with the model group, thymus and spleen indexes of the intervention groups increased, and the difference was statistically significant(P<0.05). And compared with the blank group, the contents of cytokines IL-2, IL-4 and IFN-γ of the rats in the model group decreased significantly, the contents of TNF-α raised notably, and the differences were statistically significant (P<0.05); Furthermore, compared with the model group, the contents of cytokines IL-2, IL-4 and IFN-γ of the rats in the intervention groups raised significantly, the contents of TNF-α decreased, the difference was statistically significant (P<0.05). Conclusion: Extract of selenium-enriched Astragalus membranaceus can significantly improve the immune function of immunosuppressive rat model induced by cyclophosphamide and the effects are more significant than common astraguls membranaceus.
