摘要:
目的:研究臭灵丹对人白血病K562和NB4细胞的抑制作用及其黄酮类成分。方法:应用四甲基偶氮噻唑蓝比色法(MTT法)测臭灵丹对K562和NB4细胞生长的抑制率,采用PI/Annexin-V双染色方法考察臭灵丹对K562和NB4的凋亡作用;运用大孔树脂、Sephadex LH-20柱色谱进行化学成分的分离纯化,通过波谱分析鉴定化合物结构。结果:臭灵丹对K562和NB4细胞的抑制作用呈现剂量依赖性;臭灵丹能剂量依赖性地诱导K562和NB4细胞的凋亡;从臭灵丹中分离得到了6个黄酮类化合物,分别鉴定为洋艾素、金腰乙素、5-羟基-3',4',7-三甲氧基黄酮、雷杜辛黄酮醇、芹菜素-7-O-β-D-葡糖糖苷、槲皮素。结论:臭灵丹具有抗K562和NB4细胞的作用,首次从臭灵丹中分离得到雷杜辛黄酮醇。
Abstract:
Objective: To study the inhibitory effects of ChouLingDan (Laggera pterodonta) on human leukemia K562 and NB4 cells and its flavone ingredients. Methods: MTT method was used to detect the inhibitory rate of ChouLingDan on the growth of K562 and NB4 cells, PI/Annexin-V double staining method was adopted to investigate the apoptosis effects of ChouLingDan on K562 and NB4 cells; macroporous resin and Sephadex LH-20 column chromatography were adopted to seperate and purify chemical ingredients, and spectrum was applied to analyze and identify the structure of chemical compound. Results: The inhibitory effects of ChouLingDan on K562 and NB4 cells showed in a dose-dependent manner; ChouLingDan could induce the apoptosis of K562 and NB4 cells in a dose-dependent manner; six kinds of flavanoids were separated from ChouLingDan, and they were identified as absinthin, chrysosptertin B,5-hydroxy-3', 4', 7-trimethoxyflavone, retusine, apigemin-7-O-β-D-glucopyranoside and quercetin. Conclusion: ChouLingDan could inhibit K562 and NB4 cells, and retusine is isolated from the plant initially.
