摘要:
目的:探讨髓芯减压联合生骨再造丸对激素性家兔坏死股骨头骨形成蛋白2(BMP-2)与血管内皮生长因子mRNA(VEGF mRNA)的影响。方法:将成年雄性青紫兰兔60只随机分为5组,正常对照组(N)8只,模型对照组(Y)、生骨再造丸组(M)、手术组(S)、综合治疗组(MS),每组13只。Y、M、S、MS组采用内毒素加激素联合造模,造模4周后每组随机处死1只兔子,检验造模是否成功。造模成功后N组和Y组灌服生理盐水20 mL/d,M组灌服生骨再造丸溶液4.6 mL/(kg·d),S组采用髓芯减压术治疗,MS组在手术治疗基础上灌服同量生骨再造丸溶液,各组分别干预8周,处死动物所获股骨头行HE染色,采用RT-PCR观察骨组织中BMP-2与VEGF mRNA表达情况。结果:与Y组比较,各治疗组空陷窝率下降,BMP-2和VEGF mRNA表达水平均有不同程度升高,差异有统计学意义(P<0.05),其中以MS组空陷窝率降低最为显著(P<0.01)。结论:增强股骨头骨组织BMP-2和VEGF mRNA表达是髓芯减压术联合生骨再造丸治疗激素性兔股骨头坏死的作用机制之一。
Abstract:
Objective: To discuss the impacts of core decompression and ShengGu ZaiZao pills on BMP-2 and VEGF mRNA of the rabbits suffering steroid-induced femoral head necrosis. Methods: Sixty patients were randomized into five groups, eight rabbits in normal control group as N, model control group, ShengGu ZaiZao pills group, operation group and comprehensive therapy group as Y, M, S and MS, 13 rabbits each group. The rabbits in Y, M, S and MS were prepared into the models by endotoxin and hormone, in four weeks after modeling, five rabbits in each group (N,Y, M, S and MS) were sacrificed randomly to testify whether the modeling succeeded. After successfully modelling, N and Y were drenched with physiological saline, 20 mL/d, M with the solution of ShengGu ZaiZao pills, 4.6 mL/(kg·d), S accepted core decompression, MS with the solution of ShengGu ZaiZao pills in the same volume based on the operation, different groups were intervened for eight weeks respectively, femoral head of the sacrificed rabbits were performed HE staining, the expressions of VEGF and BMP-2 mRNA in bone tissue were observed by RT-PCR.Results: Compared with Y, blank pit rate of different treatment groups decreased, the expressions of BMP-2 and VEGF mRNA raised to different degrees, and the difference had statistical meaning (P<0.05), among them, the decrease of comprehensive therapy group was the most signficant (P<0.01). Conclusion: Enhancing the expressions of VEGF and BMP-2 mRNA in bone tissue of femoral head is one of the mechanisms of core decompression and ShengGu ZaiZao pills in the treatment for steroid-induced femoral head necrosis.
